摘  要:为探究吡咯喹啉醌(pyrroloquinoline quinone,PQQ)对小鼠卵母细胞体外成熟的影响,该研究以6~8周龄体重相近的雌性昆明小鼠卵母细胞为研究对象,添加PQQ后通过分析第一极体排出率、活性氧(reactive oxygen species,ROS)水平、谷胱甘肽(glutathione,GSH)水平、线粒体膜电位(mitochondrial membrane potential,MMP)水平以及细胞内线粒体活性氧(mitochondrial reactive oxygen species,Mito ROS)水平,探讨PQQ对小鼠卵母细胞体外成熟的影响。结果表明:添加10μmol/L PQQ可极显著降低卵母细胞内ROS水平和Mito ROS水平,极显著提高卵母细胞第一极体排出率、GSH水平,显著提高MMP水平,抗氧化相关基因Sirt1、GPX3、CAT、SOD2的mRNA表达水平也有显著的上调。以此可以推测,在体外成熟过程中添加10μmol/L PQQ可以提高小鼠卵母细胞的成熟率。In order to explore the effect of pyrroloquinoline quinone(PQQ)on the in vitro maturation of mouse oocytes,the oocytes of female Kunming mice aged 6~8 weeks with similar body weights were used as the research object.PQQ was added,and the first polar body extrusion rate,the level of reactive oxygen species(ROS),the level of glutathione(GSH),the level of mitochondrial membrane potential(MMP),and the level of mitochondrial reactive oxygen species(Mito ROS)in cells were detected to analyze the effect of PQQ on the in vitro maturation of mouse oocytes.The results showed that adding 10μmol/L PQQ could significantly reduce the levels of ROS and Mito ROS in oocytes,significantly increase the first polar body extrusion rate of oocytes and the level of GSH,and significantly increase the level of MMP.The mRNA expression levels of antioxidant-related genes Sirt1,GPX3,CAT,and SOD2 were also significantly upregulated.Therefore,it can be speculated that adding 10μmol/L PQQ during the in vitro maturation process can improve the maturation rate of mouse oocytes.
