摘 要:目的分析维生素D_(3)(VD_(3))对大鼠血浆中维生素D_(2)(VD_(2))浓度的影响,为临床合理使用维生素D提供理论依据。方法采用SD大鼠构建VD_(3)补充组和VD_(3)缺乏组模型,每组6只。利用高效液相色谱-串联质谱(HPLCMS/MS)法检测血浆中25-羟基维生素D_(3)[25(OH)VD_(3)]和25-羟基维生素D_(2)[25(OH)VD_(2)]的含量,并通过独立样本t检验分析两组间25(OH)VD_(3)和25(OH)VD_(2)水平的差异。结果两组大鼠体重增加量、每日水消耗量比较,差异无统计学意义(P>0.05);VD_(3)补充组大鼠每日饲料消耗量高于VD_(3)缺乏组,差异有统计学意义(P<0.05)。HPLC-MS/MS法检测25(OH)D_(2)和25(OH)VD_(3)时线性关系良好,r均>0.990;VD_(3)补充组大鼠体内25(OH)VD_(2)水平随喂养时间降低,而25(OH)VD_(3)水平随喂养时间升高(P<0.05)。结论D_(3)的补充可降低大鼠体内25(OH)VD_(2)水平,提示不同形式维生素D在体内存在相互影响。Objective To analyze the effect of vitamin D_(3)(VD_(3))on the concentration of vitamin D_(2)(VD_(2))in rat's plasma in order to provide a theoretical basis for the rational use of vitamin D in clinic.Methods SD rats were used to construct models of the VD_(3)supplementation and the VD_(3)deficiency,with 6 rats in each group.The high performance liquid chromatographytandem mass spectrometry(HPLC-MS/MS)was employed to detect the contents of 25-dihydroxy vitamin D_(3)[25(OH)VD_(3)]and 25-dihydroxy vitamin D_(2)[25(OH)VD_(2)]in plasmas.The independent-samples t-test was used to analyze the differences in the levels of 25(OH)VD_(3)and 25(OH)VD_(2)between VD_(3)supplementation and VD_(3)deficiency groups.Results There were no statistically significant differences in the values of rats'weight gain and daily water consumption between the two groups(P>0.05).The daily feed consumption of rats in the VD_(3)supplementation group was higher than that in the VD_(3)deficiency group,and the difference was statistically significant(P<0.05).The linearities of 25(OH)VD_(2)and 25(OH)VD_(3)detected by HPLC-MS/MS method were good(r>0.990).In the VD_(3)supplementation group,25(OH)VD_(2)levels in rats decreased,while 25(OH)VD_(3)levels in rats increased with the increasing feeding time(P<0.05).Conclusions The supplementation of VD_(3)can significantly reduce the 25(OH)VD_(2)levels in rats,which suggests that different forms of vitamin D interact with each other in vivo.
