姜黄素对S.aureus引起的奶牛乳腺上皮细胞自噬、炎症和凋亡的影响

摘  要:拟探究姜黄素在金黄色葡萄球菌(Staphylococcus aureus,S.aureus)感染的奶牛乳腺上皮细胞生理中的作用。采用免疫荧光和蛋白质免疫印迹(Western blotting,WB)法,检测姜黄素对S.aureus感染后奶牛乳腺上皮细胞中细胞自噬、炎性反应和细胞凋亡的影响。结果显示,S.aureus MOI为8∶1,37℃下孵育2 h可有效的使S.aureus侵入乳腺上皮细胞,并且S.aureus能够阻止自噬泡的降解;使用姜黄素处理后,胞内自噬通量增加,主要表现在胞内Beclin-1和LC3水平上升,P62水平降低;使用姜黄素处理后,胞内促炎性因子IL-1β水平降低,抑炎性因子IL-10水平上升;与此同时使用姜黄素能够有效缓解S.aureus引起的细胞凋亡水平升高。研究结果表明,姜黄素对S.aureus引起的奶牛乳腺上皮细胞炎症有治疗作用。The aim of this study was to investigate the role of curcumin in the physiology of S.aureus-infected bovine mammary epithelial cells.The effects of curcumin on autophagy,inflammatory response and apoptosis in bovine mammary epithelial cells after S.aureus infection were detected by immunofluorescence and Western blotting(WB).The results showed that the MOI of S.aureus was 8∶1,37℃for 2 h can effectively make S.aureus invade mammary epithelial cells,and S.aureus can prevent the degradation of autophagic vacuoles;after treatment with curcumin,the intracellular autophagy flux increased,mainly manifested in the increase of intracellular Beclin-1 and LC3 levels,and the decrease of P62 level;after treatment with curcumin,the level of intracellular pro-inflammatory factor IL-1β decreased,and the level of anti-inflammatory factor IL-10 increased.At the same time,the use of curcumin can effectively alleviate the increase of apoptosis level caused by S.aureus.The results showed that curcumin had a therapeutic effect on the inflammation of bovine mammary epithelial cells caused by S.aureus.
