摘 要:目的基于网络药理学探讨鱼腥草治疗细菌性角膜炎(BK)和真菌性角膜炎(FK)的作用机制。方法通过中药系统药理学数据库分析平台(TCMSP)数据库和化学数据库获取鱼腥草的主要化学成分。通过Swiss Target Prediction数据库和TargetNet数据库获取鱼腥草化学成分的作用靶点。应用GeneCards数据库和DisGeNET数据库获取BK、FK的潜在靶点。通过STRING数据库和Cytoscape软件构建蛋白相互作用(PPI)网络和“活性成分-核心靶点-关键通路”网络。通过DAVID数据库对鱼腥草治疗BK、FK的潜在作用靶点进行基因本体(GO)富集分析和京都基因与基因组百科全书(KEGG)通路富集分析,预测其作用机制。结果研究筛选出鱼腥草主要活性成分14种,潜在靶点858个,经分析确定治疗BK的潜在靶点有70个,治疗FK的潜在靶点有52个。PPI分析结果发现,鱼腥草治疗BK的核心蛋白12个,治疗FK的核心蛋白15个。GO富集分析和KEGG通路富集分析揭示了相关生物学过程、分子功能、细胞组成及主要通路。“活性成分-核心靶点-关键通路”网络发现,槲皮素是鱼腥草治疗BK、FK的关键活性化合物;治疗BK的关键靶点为蛋白激酶B1(AKT1),治疗FK的关键靶点为丝裂原活化蛋白激酶14(MAPK14);核因子κB(NF-κB)信号通路是其共同的关键作用通路。结论鱼腥草治疗BK和FK具有多成分、多靶点、多途径协同作用的特点,槲皮素为其核心活性成分。Objective To explore the mechanism of action of Houttuynia cordata Thunb.in treating bacterial keratitis(BK)and fungal keratitis(FK)based on network pharmacology technology.Methods The main chemical components of Houttuynia cordata Thunb.were obtained from Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform(TCMSP)database and chemical database.The action targets of the chemical components of Houttuynia cordata Thunb.were obtained from Swiss Target Prediction database and TargetNet database.GeneCards database and DisGeNET database were used to obtain the potential targets of BK and FK.Protein-protein interaction(PPI)network and“active ingredient-core target-key pathway”network were constructed by using STRING database and Cytoscape software.DAVID database was used to perform Gene Ontology(GO)enrichment analysis and Kyoto Encyclopedia of Genes and Genomes(KEGG)pathway enrichment analysis on the potential targets of Houttuynia cordata Thunb.in treating BK and FK,and the mechanisms of action were predicted.Results In this study,14 main active ingredients of Houttuynia cordata Thunb.and 858 potential targets were screened out,and 70 potential targets for treating BK and 52 potential targets for treating FK were determined after analysis.The results of PPI analysis showed that 12 core proteins were related to the treatment of Houttuynia cordata Thunb.for BK and 15 core proteins were related to the treatment of Houttuynia cordata Thunb.for FK,and GO enrichment analysis and KEGG pathway enrichment analysis revealed the related biological processes,molecular functions,cell composition and main pathways.The“active ingredient-core target-key pathway”network showed that quercetin in Houttuynia cordata Thunb.was the key active compound in treating BK and FK.The key target of treating BK was protein kinase B1(AKT1),and the key target of treating FK was mitogen-activated protein kinase 14(MAPK14).The nuclear factor-kappa B(NF-κB)signaling pathway was their common key action pathway.Conclusi
