黄芩素、小檗碱、大黄酸、大黄素联合阿莫西林对幽门螺杆菌诱导GES-1分泌IL-8的影响

摘　　要：目的 研究中药活性成分——黄芩素、小檗碱、大黄酸、大黄素与阿莫西林(AMC)联用时对幽门螺杆菌(Hp)诱导胃上皮细胞(GES-1)分泌白细胞介素-8(IL-8)的影响。方法 2022年10月采用酶联免疫吸附测定法检测黄芩素、小檗碱、大黄酸、大黄素,以及与AMC联用时GES-1中IL-8的表达水平。结果 黄芩素、小檗碱、大黄酸、大黄素单独使用时均能明显降低IL-8表达水平,小檗碱、大黄素联合AMC均能明显降低IL-8表达水平,差异均有统计学意义(P<0.05);大黄素联用AMC时抑制IL-8分泌的效果最强。黄芩素联用AMC时IL-8表达水平与单独使用AMC比较,差异无统计学意义(P>0.05)。结论 小檗碱、大黄素均能通过降低Hp诱导GES-1分泌IL-8表达水平而增强AMC的抗Hp活性。黄芩素、大黄酸均不能通过降低IL-8表达水平而增加AMC的抗Hp活性。Objective To study the effect of baicalin,berberine,rhein and emodin combined with amoxicillin(AMC)on expressions of Inter leukin-8(IL-8)in GES-1 cells with the treatment of Helicobacter pylori(Hp)infection.Methods The protocol was designed by ELISA methods,determined the expressions of IL-8 in GES-1 cells pretreated with baicalin,berberine,rhein,emodin and combined with AMC against Hp infection in October,2022.Results Baicalin,berberine,rhein and emodin could reduce the expressions of IL-8.Rhein,emodin combined with AMC could reduce the expressions of IL-8(P<0.05).In the study,emodin combined with AMC has the strongest effect on the inhibition of IL-8 secretion in GES-1 cells.There was no significant difference between baicalein combined with AMC,compared with AMC alone(P>0.05).Conclusion Berberine and emodin can significantly increase the anti-Hp activity of AMC by decreasing the secretion of IL-8 in GES-1 cells.However,baicalin and rhein could not increase the anti-Hp activity of AMC by decreasing the secretion of IL-8.
