摘 要:孟德尔随机化(MR)是一种以遗传变异作为工具变量探索暴露因素与研究结局因果关系的统计学方法,可有效排除混杂因素和反向因果的干扰。近年来,MR方法随着全基因组关联研究的发展及大量汇总数据集的公开在医学各领域中已有了一定的应用。该文对MR原理及其在医学影像学中的具体应用展开了概述,通过与随机对照试验的研究进行比较,对其在医学影像学领域所面临的机遇与挑战进行了论述,以推进MR在医学影像学领域的应用。Mendelian randomization(MR)is a statistical method that uses genetic variation as instrumental variables to explore the causal relationship between exposure factors and research outcomes,which can effectively eliminate the interference of confounding factors and reverse causality.In recent years,with the development of Genome-Wide association studies and the publication of a large number of aggregated data sets,the MR method has been applied in various fields of medicine.In this paper,the principle of MR and its application in medical imaging are summarized,and the opportunities and challenges in the field of medical imaging are discussed by comparing with randomized controlled trials,in order to promote the application of MR in the field of medical imaging.
