摘  要:目的通过构建大鼠脊髓损伤(SCI)模型,探究羽扇豆醇(Lupeol)极化M1/M2巨噬细胞对SCI大鼠神经功能恢复的影响。方法采用改良Allen's法建立大鼠SCI模型,将大鼠随机分为3组,分别为假手术组(Sham组)、SCI组和Lupeol组,每组18只。Lupeol组于损伤后立刻给予Lupeol腹腔注射治疗14 d。采用免疫荧光染色观察脊髓损伤中心巨噬细胞的极化程度,采用ELISA检测脊髓组织中白细胞介素-1β(IL-1β)、肿瘤坏死因子-α(TNF-α)、IL-6水平,通过尼氏染色观察损伤中心神经元存活量,通过BBB评分和斜板试验评估大鼠肢体运动功能恢复情况。结果免疫荧光染色显示,与SCI组比较,Lupeol组损伤中心的iNOS^(+)F4/80^(+)细胞/F4/80^(+)细胞比率明显减少,而CD206^(+)F4/80^(+)细胞/F4/80^(+)细胞比率明显增加,差异均有统计学意义(t=4.847、4.856,P均<0.05)。ELISA提示,与SCI组相比,Lupeol组和Sham组脊髓组织中IL-1β、TNF-α和IL-6水平均显著降低,差异均有统计学意义(F=92.49、130.30、69.52,P均<0.05)。尼氏染色显示,与SCI组相比,Lupeol组和Sham组损伤中心的神经元数量显著增加,差异有统计学意义(F=23.01,P<0.05)。BBB评分显示,SCI组和Lupeol组的运动功能在SCI后第3天差异无统计学意义(P>0.05)。从第7天开始,接受Lupeol治疗大鼠的运动评分显著高于SCI组,差异均有统计学意义(P均<0.05)。斜板试验显示,与SCI组相比,Lupeol组和Sham组大鼠的倾斜角度均显著升高,差异有统计学意义(F=80.33,P<0.05)。结论Lupeol可能通过极化M1/M2巨噬细胞来减少损伤中心的炎症反应,改善局部微环境,减少神经元凋亡,有效促进大鼠的肢体运动功能恢复。Objective To investigate the effects of Lupeol(Lupeol)polarized M1/M2 macrophages on the recovery of neurological function in rats with spinal cord injury(SCI).Methods The rat SCI model was established by modified Allen's method,and the rats were randomly divided into three groups:sham operation group(Sham group),SCI group and Lupeol group,18 per group.Lupeol group was treated with intraperitoneal injection of Lupeol immediately after injury for 14 days.Immunofluorescence staining was used to observe the polarization of macrophages in the center of spinal cord injury.ELISA was used to detect the levels of interleukin⁃1β(IL⁃1β),tumor necrosis factor⁃α(TNF⁃α)and interleukin⁃6(IL⁃6)in spinal cord tissue.Nissl staining was used to observe the survival of neurons in the injured center.Basso-Beattie-Bresnahan(BBB)score and inclined plate test were used to evaluate the recovery of limb motor function.Results Immunofluorescence staining showed that the ratio of iNOS^(+)F4/80^(+)cells/F4/80^(+)cells in the injury center of the Lupeol group was significantly reduced,while the ratio of CD206^(+)F4/80^(+)cells/F4/80^(+)cells was significantly increased,and the difference was statistically significant(t=4.847,4.856,all P<0.05).ELISA showed that compared with the SCI group,the levels of IL⁃1β,TNF⁃αand IL⁃6 in the spinal cord tissue of the Lupeol group and Sham group were significantly lower(F=92.49,130.30,69.52;all P<0.05).Nissl staining showed that compared with the SCI group,the number of neurons in the injury center of the Lupeol group and Sham group increased significantly(F=23.01,P<0.05).The BBB score showed that there was no statistically significant difference in motor function between the SCI group and the Lupeol group on the 3rd day after SCI(P>0.05).Starting from the 7th day,the motor scores of rats treated with Lupeol were significantly higher than those of the SCI group(all P<0.05).The inclined plate test showed that compared with the SCI group,the tilt angles of the rats in the Lupeol group
