摘 要:目的 通过检测长链非编码RNA(lncRNA)核仁小分子RNA宿主基因6(SNHG6)、微小RNA-186 (miR-186)在侵袭性垂体腺瘤(IPA)中的表达变化,并分析其临床意义。方法 收集河北省邯郸市中心医院收治的105例垂体腺瘤患者为研究对象,将其分为非侵袭组(54例)和侵袭组(51例);比较两组SNHG6、miR-186表达水平及其与临床特征及预后的关系。用Pearson相关系数分析SNHG6与miR-186的相关性。结果 侵袭组腺瘤组织SNHG6表达水平(2.57±0.48)显著高于非侵袭组(1.00±0.11),miR-186表达水平(0.49±0.12)显著低于非侵袭组(1.01±0.08),差异均有统计学意义(均P<0.05)。IPA患者不同年龄、性别、肿瘤分型中SNHG6、miR-186表达水平比较,差异均无统计学意义(均P>0.05),而在不同肿瘤体积大小比较差异均有统计学意义(均P<0.05)。IPA患者腺瘤组织SNHG6与miR-186表达水平呈负相关(r=-0.578,P=0.000)。SNHG6低表达患者复发率(12.0%)显著低于SNHG6高表达患者(50.0%),差异有统计学意义(Logrankχ^(2)=8.913,P=0.003)。miR-186高表达患者的复发率显著低于低表达患者(15.4%对比48.0%),差异有统计学意义(Logrankχ^(2)=6.452,P=0.011)。结论 IPA腺瘤组织中SNHG6表达上调,miR-186表达下调,SNHG6与miR-186呈负相关,两者均与肿瘤体积大小及预后相关。Aim To detect the expression changes of long non-coding RNA(lncRNA)nucleolar small RNA host gene(SNHG6)and microRNA-186(miR-186)in invasive pituitary adenoma(IPA)tissue,and analyze their clinical significance.Methods A total of 105 patients with pituitary adenoma treated in Handan Central Hospital,Hebei Province were studied,and were separated into a non-invasive group(54 cases)and an invasive group(51 cases).The expression levels of SNHG6 and miR-186 were compared between the two groups.The relationship between the expression levels of SNHG6 and miR-186 in adenoma tissues and clinical features and prognosis was analyzed.The correlation between SNHG6 and miR-186 was analyzed by Pearson correlation coefficient.Results The expression level of SNHG6 in adenoma tissue of the invasive group(2.57±0.48)was obviously higher than that of the non-invasive group(1.00±0.11),and the expression level of miR-186(0.49±0.12)was obviously lower than that of the non-invasive group(1.01±0.08)(all P<0.05).There were no significant differences in the expression levels of SNHG6 and miR-186 in IPA patients with different ages,genders and tumor types(P>0.05),but there were significant differences in different tumor sizes(P<0.05).The expression levels of SNHG6 and miR-186 in adenoma tissues of patients with IPA were negatively correlated(r=-0.578,P=0.000).The recurrence rate of patients with low SNHG6 expression(12.00%)was obviously lower than that of patients with high SNHG6 expression(50.00%)(Logrankχ^(2)=8.913,P=0.003).The recurrence rate of miR-186 with high expression(15.4%vs.48.00%)was significantly lower than that of patients with low expression(Logrankχ^(2)=6.452,P=0.011).Conclusion The expression of SNHG6 is up-regulated and the expression of miR-186 is down-regulated in IPA adenoma tissue.SNHG6 is negatively correlated with miR-186,and both are related to tumor size and prognosis.
