摘 要:目的建立痰浊血瘀证ED大鼠模型,筛查ED病证大鼠主动脉差异表达蛋白质并进行生物信息学分析,为研究痰浊血瘀证ED发病机制提供新思路。方法将120只雄性性成熟SD大鼠随机分为正常对照组(N组,n=10)和痰浊血瘀证建模组(M组,n=110),分别以正常饲料和高脂饲料饲养,全程动态观测生物学表征、阴茎勃起功能与性行为学改变,饲养8周建模组至成功建立痰浊血瘀证候模型后,以阴茎勃起实验和性行为学实验从中筛选ED大鼠,并随机取10只设为ED病证组(其余ED病证大鼠与证候大鼠用于其他研究),取材后全自动生化分析仪检测血脂四项,全自动血液流变仪检测血流变。TMT技术鉴定ED组大鼠主动脉组织差异蛋白质并进行GO与KEGG通路分析。结果(1)模型建立:与N组相比,M组大鼠的体重及核心温度显著增加(P<0.001),饮食量、饮水量、尿量和粪便量显著减少(P<0.001)。(2)阿扑吗啡阴茎勃起与性行为学实验:与N组相比,M组大鼠勃起、爬背、插入和射精伏期显著延长(P<0.001),勃起次数、爬背次数、插入次数和射精次数显著减少(P<0.001),ED成模率为72.7%。(3)血脂四项与血液流变学指标:与N组相比,ED组大鼠血清中TC、TG和LDL-C含量显著升高(P<0.05),HDL-C含量显著降低(P<0.05)。与N组相比,ED组大鼠的全血黏度、血浆黏度、红细胞压积和血沉方程K值均显著升高(P<0.05)。(4)蛋白质组学:共鉴定出痰浊血瘀证ED大鼠动脉相关差异表达蛋白质200种(上调121种,下调79种),GO分析显示差异蛋白质主要参与血管调节过程、脂质氧化代谢和脂肪酸氧化代谢等过程。KEGG信号通路分析发现差异蛋白质涉及雌激素信号通路、氨基酸生物合成通路和碳代谢通路等114条信号通路。结论(1)高脂饮食可成功构建痰浊血瘀证ED大鼠模型,其生物学表征、血脂四项及血液流变学改变特征与中医临床证候表现相吻合;(2)痰浊血瘀证ED大鼠动�Objective Establish a rat model of phlegm turbidity and blood stasis ED syndrome,screen for differentially expressed proteins in the aorta of ED syndrome rats,and conduct bioinformatics analysis to provide new ideas for studying the pathogenesis of phlegm turbidity and blood stasis ED.Methods 120 male sexually mature SD rats were randomly divided into a normal control group(N group,n=10)and a phlegm turbidity and blood stasis modeling group(M group,n=110),feeding with normal feed and high-fat feed respectively,the biological characteristics,sexual behavior,and penile erection function were dynamically observed throughout the process.After 8 weeks of feeding until a successful model of phlegm turbidity and blood stasis syndrome was established,ED rats were selected through penile erection experiments and sexual behavior experiments,and 10 rats were randomly selected as the ED disease group(the remaining ED rats and syndrome rats were used for other studies).After sampling,four blood lipid parameters were detected by a fully automated biochemical analyzer,and blood rheology was detected by a fully automated blood rheometer.TMT technology identification of differential proteins in the aortic tissue of ED group rats and GO analysis and KEGG pathway analysis.Results(1)Model establishment:Compared with group N,group M rats showed a significant increase in body weight and core temperature(P<0.001),as well as a significant decrease in diet,water,urine,and feces(P<0.001).(2)APO Penile Erection and Sexual Behavior Experiment:Compared with group N,group M rats showed significantly prolonged periods of erection,back climbing,intromssion,and ejaculation(P<0.001),and significantly reduced frequency of erection,back climbing,insertion,and ejaculation(P<0.001).The ED modeling rate was 72.7%.(3)Blood lipids and hemorheology:Compared with group N,the levels of TC,TG,and LDL-C in the serum of rats in the ED group were significantly increased(P<0.05),while the level of HDL-C was significantly decreased(P<0.05).Compared with group N,
