摘 要:目的探究雄激素剥夺对2型糖尿病(T2DM)小鼠糖脂代谢的影响。方法采用高脂饮食(HFD)联合链脲佐菌素(STZ)构建T2DM小鼠模型。研究对象分为四组:对照组(CON)、T2DM组(T2DM)、T2DM+睾丸切除组(T2DM-ORC)和T2DM+睾丸切除+比卡鲁胺组(T2DM-ORC-BIC)。干预后,记录小鼠体重和生理生化指标,并观察脏器病理变化。结果雄激素剥夺显著升高了血清胰岛素水平(P<0.05),并降低了HDL-C(P<0.05)。同时,空腹血糖水平和胰岛素抵抗指数增加,加重了葡萄糖和胰岛素耐量,导致血清TC、TG、LDL-C及NEFA水平显著升高。结论雄激素剥夺从体重、脏器病理损伤和糖脂代谢多方面加重了T2DM小鼠的代谢紊乱,为临床上雄激素剥夺与T2DM病理变化的关系提供了理论基础和实验支持。Objective To investigate the effects of androgen deprivation on glucose and lipid metabolism in type 2 diabetes mellitus(T2DM)mice.Methods T2DM mouse models were developed by high-fat diet(HFD)combined with streptozotocin(STZ),and research object were divided into 4 groups including the control group(CON),T2DM group(T2DM),T2DM+orchiectomy group(T2DM-ORC)and T2DM+orchiectomy+bicalutamide group(T2DM-ORC-BIC).After the intervention,the changes in the body weight and physiological/biochemical indexes of mice were recorded,and the pathological changes of organs were observed.Results Androgen deprivation significantly elevated the level of serum insulin(P<0.05)and decreased the level of serum HDL-C(P<0.05).Simultaneously,fasting blood glucose and insulin resistance index were also increased,which aggravated glucose and insulin tolerance,resulting in significant increases in the levels of serum TC,TG,LDL-C and NEFA.Conclusion Androgen deprivation aggravates metabolic disorders in T2DM mice from weight,organ pathological injury and glucose and lipid metabolism,which provides theoretical basis and experimental support for the clinical relationship between androgen deprivation and T2DM.
