摘 要:目的构建和评判慢性睡眠限制(Chronic sleep restriction,CSR)诱导雄性性腺功能减退(Male hypogonadism,MH)大鼠模型。方法30只Wistar大鼠按随机区组法分为对照(Control,CON)组、急性睡眠限制(Acute sleep restriction,ASR)组、慢性睡眠限制(CSR)组,其中ASR组和CSR组进行每天长达20 h睡眠限制干预,分别持续3 d和35 d,CON组正常饲养。完成实验后,进行交配实验。称取睾丸重量,计算睾丸指数,检测血清T水平。结果与CON组相比,ASR组的血清T、性功能、睾丸指数无明显变化;CSR组血清T水平下降(P<0.01),并伴有活跃度下降、性功能减退(P<0.01)、睾丸指数降低(P<0.01)等改变。结论本研究利用慢性睡眠限制方法(20 h/d,35 d)构建的MH大鼠模型具有可行性。Objective To construct and evaluate a rat model of male hypogonadism(MH)induced by chronic sleep restriction.Methods Thirty Wistar rats were randomly divided into the control group(CON),acute sleep restriction group(ASR),and chronic sleep restriction(CSR)group.The rats in the ASR and CSR groups were subjected to sleep restriction interventions,and the intervention respectively lasted for 3 and 35 days,and 20 h each day.The rats in the CON group were reared and slept normally.After completion of the experiment,mating experiment was performed.Their testes were collected and weighed,testicular index was calculated and serum T levels were measured by ELISA.Results No significant changes were found in serum T,sexual function,and testicular index in the ASR group compared with those in the CON group;However,in the CSR group,serum T level(P<0.01),signs and symptoms such as fatigue,sexual function decline(P<0.01),and testicular index(P<0.01)were all prominently declined.Conclusion Our results suggest that it is feasible to construct MH model using chronic sleep restriction method(20 h/d,35 d).
