摘 要:目的观察翘芍方对早泄大鼠模型的干预效果,并探讨其作用机制。方法根据早泄连续统一体理论筛选36只早泄大鼠模型,随机分为4组,空白组、模型组生理盐水灌胃,翘芍方组10 g·kg^(-1)·d^(-1)翘芍方灌胃,阳性对照组0.6 mg/kg盐酸达泊西汀同笼前3 h灌胃。第4周记录大鼠30 min交配行为。取下丘脑组织,ELISA测5-HT含量,PT-PCR测TPH2、5-HTT、5-HT1A、5-HT2C表达。结果模型组的IL、EL、MF明显减少,EF明显增加,下丘脑5-HT含量、TPH2 mRNA明显减少,5-HTT mRNA、5-HT1A受体mRNA明显增加(P<0.01);与模型组相比,翘芍方组EL、MF、IF明显增加,EF明显减少,下丘脑5-HT含量、TPH2 mRNA、5-HT2C受体mRNA明显增加,5-HTT mRNA、5-HT1A受体mRNA明显减少(P<0.01)。结论翘芍方可延长早泄大鼠射精潜伏期,其机制可能与促进5-HT合成、抑制5-HT转运、调控5-HT受体活性有关。Objective To evaluate the intervention effect of Qiaoshao formula on rat models with premature ejaculation,and explore its molecular mechanism.Methods Based on the theory of continuous unity of premature ejaculation,36 rat models with premature ejaculation were selected and randomly divided into a blank group and a model group,the rats in the blank group were given with physiological saline by gavage,the rats in the Qiaoshao formula group were orally administrated with 10 g·kg^(-1)·d^(-1)Qiaoshao formula.The rats in the positive control group were given with 0.6 mg/kg dapoxetine hydrochloride by gavage 3 hours before cage placement.Record the mating behavior of rats for 30 minutes in the fourth week.Take the hypothalamus,measure the 5-HT content by ELISA,and measure the expression of TPH2,5-HTT,5-HT1A,and 5-HT2C by PT-PCR.Results The IL,EL,and MF levels of rats in the model group were significantly decraesed,while EF was significantly increased.Meanwhile,hypothalamic 5-HT content and TPH2 mRNA level were significantly reduced,while 5-HTT mRNA and 5-HT1A receptor mRNA were significantly increased(P<0.01).Compared with those of the control group,EL,MF,and IF levels of the Qiaoshao formula group were increased significantly,but EF was decreased significantly.The hypothalamic 5-HT content,TPH2 mRNA,and 5-HT2C receptor mRNA levels were increased significantly,but 5-HTT mRNA and 5-HT1A receptor mRNA levels were decreased significantly(P<0.01).Conclusion Qiaoshao formula can prolong the latency period of ejaculation in premature ejaculation rats,and its mechanism may be related to 5-HT synthesis increase,5-HT transport inhibition,and 5-HT receptor activity.
