健阳片对雄性大鼠生殖系统氧化应激损伤的调控机制研究

摘　　要：目的探讨健阳片对镉诱导雄性大鼠生殖损伤的影响与调控机制。方法使用氯化镉构建生殖损伤大鼠模型。雄性大鼠分为正常对照组、模型对照组、五子衍宗丸组及健阳片低(0.48 g/kg,2倍等效剂量)、中(0.96 g/kg,4倍等效剂量)、高剂量组(1.92 g/kg,8倍等效剂量),给药56 d。计算性腺指数及附性腺指数,检测精液参数、睾丸组织谷胱甘肽过氧化物酶(GSH-PX)、超氧化物歧化酶(SOD)、丙二醛(MDA)、血清卵泡刺激素(FSH)、黄体生成素(LH)、睾酮(T)水平,观察性腺及附性腺病理组织学改变。结果与模型对照组相比,健阳片中、高剂量组的各项检测指标差异均具有统计学意义(P<0.05),且健阳片高剂量组差异更明显(P<0.01);健阳片能改善生殖损伤模型大鼠的病理状态,其疗效与剂量之间存在量-效关系。结论健阳片能够改善雄性大鼠性腺及附性腺功能、精液参数、性激素水平,改善生殖系统氧化应激状态,修复生精功能。Objective To explore the effect of Jianyang tablets on cadmium-induced reproductive injury in male rats and its regulatory mechanism.Methods Cadmium chloride was used to construct a rat model of reproductive injury.Male SD rats were divided into the normal control group,the model control group,Wuzi Yanzong pill group,and low(0.48 g/kg,2 times the equivalent dose),medium(0.96 g/kg,4 times the equivalent dose),and high dose(1.92 g/kg,8 times the equivalent dose)groups of Jianyang tablets,and the rats were administered with the drug for 56 days.The index of gonadal and accessory sex glands was calculated.Semen parameters,the level of testicular tissue glutathione peroxidase(GSH-PX),superoxidedismutase(SOD),malondialdehyde(MDA),serum follicle-stimulating hormone(FSH),luteinizing hormone(LH),and testosterone(T)were measured,and the pathological changes of gonadal and paragonadal were observed.Results Compared with the model control group,the differences in indicators of detection between the high-and medium-dose groups of Jianyang tablets were statistically significant(P<0.05),and the differences were more obvious in the high-dose group of Jianyang tablets(P<0.01);Jianyang tablets improved the pathological state of the model rats of reproductive injury,and there was a dependent relationship between the efficacy and dose.Conclusion Jianyang tablets can improve the index of gonadal and accessory sex glands,semen parameters and sex hormone level,resist the oxidative stress of the reproductive system and repair the spermatogenic function of male rats.
