摘  要:植物海藻糖-6-磷酸合酶(trehalose-6-phosphate synthase,TPS)在植物生长发育和逆境响应中具有重要意义。为了研究南方速生树种黄梁木中TPS的生物学功能,本研究从黄梁木基因组数据库中筛选到12个TPS基因,分析了其家族进化关系以及序列特征,并检测基因的表达模式。结果显示NcTPS基因随机分布于8条染色体和1个scaffold上,分为Ⅰ类和Ⅱ类TPS;NcTPS基因长度为2352~25100 bp,其中Ⅰ类TPS有14~18个外显子,Ⅱ类TPS有2~5个外显子。除NcTPS7c外,其他蛋白均有TPS和TPP(trehalose-6-phosphate phosphatase,海藻糖-6-磷酸磷酸酶)结构域。NcTPS编码区长度在1626~2847 bp之间,蛋白序列长度为541~948 aa。NcTPS蛋白除NcTPS7c显碱性外其余皆显酸性,均为亲水性蛋白。分析NcTPS的表达模式,发现NcTPS基因在黄梁木中的表达具有时空特异性,其中NcTPS11a、NcTPS11b、NcTPS7a和NcTPS7b在根中表达量较高,推测其对黄梁木根的生长发育具有重要意义。本研究为后续深入研究黄梁木TPS基因对其生长发育以及逆境响应的作用提供参考。Plant trehalose-6-phosphate synthase(TPS)plays an important role in plant growth and stress response.In order to study the biological function of TPS in the southern fast-growing tree,12 TPS genes were screened from the genome database of Neolamarckia cadamba,their family evolutionary relationship and sequence characteristics were analyzed,and gene expression patterns were detected.The results showed that the NcTPS genes were randomly distributed on eight chromosomes and one scaffold,divided into two classes named class I and class II.The length of NcTPS genes ranged from 2352 to 25100 bp,containing 14 to18 exons in class I and 2to 5 exons in class II.In addition to NcTPS7c,all other proteins have TPS and trehalose-6-phosphate phosphatase(TPP)domains.The coding regions ranged from 1626 to 2847 bp in length,and accordingly the proteins were between 541 and 948 aa.All Nc TPS proteins were hydrophilic except NcTPS7c,which showed alkalinity.Furthermore,the expression pattern of NcTPS was analyzed using transcriptome data.The result indicated that the expression of NcTPS genes was spatiotemporally specific in N.cadamba,among which NcTPS11a,NcTPS11b,NcTPS7a,and NcTPS7b were highly expressed in the roots,implying that they might have important roles in the root growth and development in N.cadamba.This study provides a reference for the subsequent in-depth study of the role of TPS genes in the growth and stress response of N.cadamba.
