摘 要:WRKY转录因子在植物生长发育、代谢以及生物胁迫和非生物胁迫等生理生化反应中充当着非常重要的角色。本研究基于向日葵基因组数据库中13个可能具有抗病功能的WRKY转录因子进行生物信息学分析,并通过RT-qPCR测定各基因在向日葵抗感锈病品种的表达量。结果表明:13个HaWRKY基因编码196~590个氨基酸,理论等电点的范围为5.3~9.65,均为疏水性蛋白且都不具有信号肽,其中只有4个HaWRKY蛋白具有跨膜结构域。亚细胞定位预测HaWRKY31、HaWRKY50、HaWRKY57定位在细胞质,HaWRKY33定位在叶绿体,其余蛋白预测结果都位于细胞核。除HaWRKY31外,剩余蛋白都含有丝氨酸(Ser)、苏氨酸(Thr)和酪氨酸(Tyr)磷酸化位点。系统进化树显示,有6个WRKY基因与已知抗生物胁迫基因同源性较高。RT-qPCR结果显示,相比于对照,13个HaWRKY基因在接种锈菌的抗病品种中表达量均上调,同时有5个基因在感病品种中的表达量出现了明显的下调,推测这些基因都有可能参与了向日葵的抗病反应。研究结果为进一步进行WRKY转录因子的功能验证以及植物抗生物胁迫的研究提供分子基础。WRKY transcription factors play a very important role in physiological and biochemical reactions like plant growth and development,metabolism,biotic and abiotic stress.Based on the Helianthus annuus genome database,this study performed a bioinformatics analysis of 13 possible WRKY transcription factors,and determined the expression of these genes in rust-resistant and-susceptible sunflower varieties by RT-qPCR.The results showed that the 13 HaWRKY genes encoded 196~590 amino acids,and the isoelectric point ranged from 5.3~9.65,all HaWRKY proteins were hydrophobic proteins and had no signaling peptides,and only 4 HaWRKY proteins had transmembrane domains.Subcellular localization predicted that Ha WRKY31,HaWRKY50,HaWRKY57 located in the cytoplasm,HaWRKY33 located in the chloroplast and others were predicted to be located in the nucleus.Except for HaWRKY31,all the other proteins have Serine(Ser),Threonine(Thr),and Tyrosine(Tyr)phosphorylation sites.6WRKY genes had higher homology with known genes related to biotic stress through the phylogeneic tree.RTqPCR results showed that the expression levels of 13 HaWRKY genes in resistant variety inoculated with rust pathogen were significantly higher than those in control,at the same time,the expression levels of 5 HaWRKY genes in susceptible variety were significantly down-regulated,suggesting that these genes may be involved in sunflower disease resistance.The results of this study provide a molecular basis for further functional validation of WRKY transcription factors and studies on plant resistance to biotic stress.
