摘 要:苎麻[Boehmeria nivea(L.)Gaudich.]生殖生长对其纤维发育有显著的抑制作用,且苎麻开花受乙烯调控。AP2/ERF转录因子在植物中对乙烯信号响应和调控生长发育具有关键作用。本研究根据苎麻转录组数据筛选到7个不同的ERF基因,在分析其在不同种质不同部位的时空表达情况基础上,克隆了一个与花芽发育密切相关的ERF基因,并进行生物信息学特征分析。结果表明:7个ERF基因在不同种质和不同组织中均表达,但表达量差异显著,存在基因型和组织器官特异性。其中Unigene9156和Unigene16687在所有种质中相对表达量较高,尤其在0.5~1.0 cm花芽(雌花芽和雄花芽)中表达量极高。Unigene9156在‘GBN08’种质0.5~1.0 cm雄花芽中的表达量是在‘GBN08’麻骨中表达量的650多倍,而在‘GBN09’种质0.5~1.0 cm雌花芽中的表达量是在‘GBN08’麻骨中表达量的338倍。因此可推测Unigene9156在调节苎麻开花和花芽发育方面可能有重要作用。以苎麻转录组数据中的Unigene9156为模板,克隆Unigene9156基因ORF,长为1302 bp,命名为BnERF1(登录号:MZ540911)。该基因编码433个氨基酸,分子质量为47.05 kD,理论等电点为9.04,含有AP2结构域的YRG和RAYD基序,是无信号肽蛋白,亚细胞定位在细胞核内。系统进化树显示苎麻与川桑、异色山黄麻、大麻的ERF蛋白同源关系较近。该研究为进一步揭示AP2/ERF调控苎麻开花提供了科学依据。The reproductive growth of ramie[Boehmeria nivea(L.)Gaudich.]significantly inhibited its fiber development,and the flowering of ramie was regulated by ethylene.AP2/ERF transcription factor plays a key role in ethylene signaling response and growth regulation in plants.In this study,7 ERF genes were screened according to the transcriptome data of ramie.Based on the analysis of their temporal and spatial expression in different parts of different germplasm,AP2/ERF genes closely related to flower bud development were cloned and analyzed by bioinformatics.The results showed that 7 ERF genes were expressed in different germplasms and tissues,but the expression levels were significantly different,and there were genotype and organ specificity.Unigene9156 and Unigene16687were highly expressed in all germplasms,especially in 0.5~1.0 cm flower buds(female and male).The expression level of Unigene9156 in 0.5~1.0 cm male flower bud of'GBN08'germplasm was 650 times higher than that in ramie bone of'GBN08',while that in 0.5~1.0 cm female flower bud of'GBN09'germplasm was 338 times higher than that in ramie bone of'GBN08'.Therefore,Unigene9156 may play an important role in regulating flowering and flower bud development of ramie.The ORF of Unigene9156,with a length of 1302 bp,was cloned from ramie transcriptome data and named BnERF1(GenBank accession number:MZ540911).The gene encoded 433 amino acids with a molecular weight of 47.05 kD and a theoretical pI of 9.04.It contained YRG and RAYD motifs of AP2 domain.It had no signal peptide and subcellular localization in the nucleus.The phylogenetic tree showed that the ERF protein of ramie had close homology with morus notabili,trema orientalis and hemp.This study provided a scientific basis for further revealing the regulation of AP2/ERF on ramie flowering.
