摘  要:为了解拟南芥中AMAR1及其旁系同源蛋白的蛋白特征,并分析它们在生长和逆境中的表达情况,本研究通过生物信息学和qPCR等方法分析了AMAR1家族蛋白特征及其表达特征。首先通过生物信息学分析在拟南芥中共鉴定了AMAR1的17个旁系同源蛋白(Paralogs),并将其命名为AMAR1家族蛋白。AMAR1家族蛋白分为Ⅰ类和Ⅱ类,其中Ⅰ类包含4个成员,Ⅱ类进一步分为Ⅱa和Ⅱb小类,分别包含5和9个成员。大多数AMAR1家族蛋白定位在叶绿体中,其中5个(AT1G15490,AT3G55880,AT1G52750,AT2-G40095和AMAR1)具有典型的跨膜结构域。大多数基因启动子区中含有脱落酸和光响应元件。结合转录组数据和qPCR分析发现,多个AMAR1基因家族成员参与非生物与生物胁迫响应应答。其中AT5G19850、AT2G26750、AT2G26740、AT4G37150、AT5G38520和AT4G24160在多种非生物胁迫处理时表达上升,而AMAR1、AT3G10840、AT4G12830、AT5G38520、AT3G05600和AT1G15490等6个基因在多种非生物胁迫时表达下降。病原细菌接种后AT2G40095、AT3G55880、AT4G24160、AT2G26740、AT1G74640、AT2G26750、AT4G37150和AMAR1的表达量显著上升,参与植物的防御响应。以上结果表明AMAR1家族蛋白参与多种胁迫应答响应,在拟南芥应对逆境和抵抗病原菌入侵过程中起重要作用。本研究为进一步探索α/β水解酶家族在植物生长发育、胁迫应答中的作用提供理论基础。In order to understand the protein characteristics of AMAR1 and its paralogous proteins in Arabidopsis thaliana,and analyze their expression in growth and stress in Arabidopsis thaliana,we systemically analyzed the property of AMAR1 and its paralogous proteins by bioinformatics and qPCR.A total of 17 paralogs of AMAR1were predicated in A.thaliana by bioinformation analysis and were named as AMAR1 family.The AMAR1 protein family is divided into ClassⅠand ClassⅡ,with ClassⅠcontaining 4 members.ClassⅡis further divided into subclassesⅡa andⅡb,containing 5 and 9 members,respectively.Most AMAR1 family proteins are predicted to be localized in chloroplasts,and five of them(AT1G15490,AT3G55880,AT1G52750,AT2G40095,and AMAR1)have typical transmembrane domains.The promoter regions of most genes contain abscisic acid and photo responsive elements.Combining transcriptome and qPCR analysis,most AMAR1 gene family members were found to be involved in abiotic and biotic stress response.AT5G19850,AT2G26750,AT2G26740,AT4G37150,AT5G38520and AT4G24160 expressed upon a variety of abiotic stress treatments.While the expression of AMAR1,AT3G10840,AT4G12830,AT5G38520,AT3G05600 and AT1G15490 decreased during a variety of abiotic treatments.The expression levels of AT2G40095,AT3G55880,AT4G24160,AT2G26740,AT1G74640,AT2G26750,AT4G37150 and AMAR1 significantly increased after inoculation of pathogenic bacteria,indicating that these genes may regulate plant defense response.Our data suggest that the AMAR1 family may be involved in a variety of stress responses and may plays an important role in environmental adaptation and resistance to pathogenic invasion of A.thaliana.Our study provides a theoretical basis for further exploring the roles ofα/βHydrolases in plant growth,development and stress response.
