分子选择结合回交转育培育转基因抗虫水稻‘吉科稻早1(cry1C*)’

摘  要:为了培育适应于吉林省中早熟稻区的抗螟虫水稻品种,以水稻‘HD1’为cry1C*及bar基因供体,以‘吉科稻早1’为受体品种,利用cry1C*基因内分子标记及耐Basta性进行前景选择,利用37个SSR标记进行背景选择,结合回交转育创制‘吉科稻早1(cry1C*)’导入系。对导入系进行Bt蛋白质含量检测、抗虫性、耐冷性、抗稻瘟病性和田间农艺性状鉴定,选择抗虫版的水稻‘吉科稻早1’。通过对供体亲本和受体亲本杂交、4次回交、2次自交的各世代水稻材料进行分子选择,创制了携带cry1C*及bar基因、检测分子标记遗传背景恢复率100%的‘吉科稻早1(cry1C*)’导入系3个(导入系1,导入系2和导入系3)。其中的导入系1叶片Bt蛋白质含量及抗虫性与供体类似,孕穗期耐冷性、叶瘟及穗颈瘟抗性、产量构成因子及产量、稻米碾米品质及外观品质、稻米食味品质等与受体类似。转基因抗虫水稻‘吉科稻早1(cry1C*)’基本培育成功。The objective was to develop rice varieties with resistance to stem borer in the medium early maturing rice area of Jilin Province.Rice introgression lines of'JKDZ1(cry1C*)'were created through backcrossing with'HD1'as cry1C*and bar gene donor and'JKDZ1'as receptor variety.The intragenic molecular marker of cry1C*and Basta resistance were used for foreground selection and 37 SSRs were deployed to make background selection during creating introgression lines.The insect-resistant edition of rice'JKDZ1'was selected based on Bt protein content of leaf blade,insect resistance,cold tolerance,blast resistance and field agronomic characteristics of introgression lines.Three introgression lines(introgression line 1,introgression line 2,introgression line 3)of rice'JKDZ1(cry1C*)'with cry1C*and bar gene and checked SSRs background recovery rate 100%were developed by molecular selection of breeding materials in each generation of F1,four backdeliveries and two selfing from donor parent and receptor parent.There were no significant difference in Bt protein content of leaf blade and insect resistance between introgression line 1 of the three lines and the donor,and no significant difference in cold tolerance at booting stage,leaf and panicle blast resistance,yield component factors and yield,rice milling and appearance quality,taste quality between introgression line 1 and the receptor.It was basically successful breeding for the transgenic insect-resistant rice'JKDZ1(cry1C*)'.
