摘  要:大豆胞囊线虫SCN(soybean cyst nematode,Heterodera glycines)是大豆生产育种实践中最重要的病害之一。抗线虫大豆品种的培育和应用推广是防治SCN的有效措施。本研究将来源于甜菜(Beta vulgaris)的NBS-LRR(nucleotide-binding site leucine-rich repeat)类抗性基因BvcZR-3和BvHs1^(pro-1)导入栽培大豆品种‘Williams 82’中。结果表明,接种胞囊线虫3号生理小种的T4和T5代转基因大豆,平均胞囊数量为1.71~12.40个/株,雌虫指数为1.84%~14.33%,均显著低于受体品种(平均胞囊数量为20.770~17.625个/株,雌虫指数为20.38%~22.35%),且在SCN寄生过程中,两个外源抗性基因的表达量显著升高,且在没接种SCN条件下,转基因大豆在叶形、株高、节数、花色等农艺性状方面与非转基因大豆品种没有显著差异。本试验研究结果表明,过表达甜菜BvcZR-3和BvHs1^(pro-1)基因显著增强了转基因大豆对SCN抗性,为进一步拓宽大豆抗性遗传基础及加快抗线虫大豆品种选育提供依据。Soybean cyst nematode SCN(Heterodera glycines)is one of the most important diseases in soybean production and breeding.The effective measure to control SCN is the cultivation and application of nematode resistant soybean varieties.In this study,NBS-LRR(nuclear binding site leucine rich repeat)resistance genes BvcZR-3 and BvHs1^(pro-1)from sugar beet were(Beta vulgaris)introduced into cultivated soybean variety'Williams 82'.The results showed that the average number of cysts of T4and T5transgenic soybean inoculated with race 3 of SCN was 1.71~12.40 per plant,and the female index was 1.84%~14.33%,which were significantly lower than those of recipient varieties(the average number of cysts was 20.770~17.625 per plant,and the female index was 20.38%~22.35%),and the expression of two foreign resistance genes increased significantly during SCN parasitism.Without SCN inoculation,there was no significant difference between transgenic soybean and non-transgenic soybean in leaf shape,plant height,number of nodes,flower color and other agronomic characters.The results demonstrated that overexpression of BvcZR-3 and BvHs1^(pro-1)significantly enhanced resistance to soybean cyst nematode in transgenic soybean plants,thus provided an alternative resistance strategy to broaden the SCN resistance background and breeding of SCN-resistant soybean cultivars.
