摘  要:杨树繁殖快,易栽培,目前已广泛用于生态防护林和工业用材林。ERF(ethylene responsive factor)转录因子是AP2/ERF家族中最大的亚族,在调节植物生长发育、应答各种非生物胁迫方面发挥着重要的作用。本研究旨在揭示ERF转录因子家族成员RAP2L15在调控杨树生长发育方面的功能。本研究以‘717杨’为材料,克隆ERF转录因子RAP2L15基因的总cDNA序列,进行植物过表达载体构建。结果表明,通过农杆菌介导法进行杨树遗传转化,获得杨树过表达转基因株系14个。RT-qPCR结果显示,与非转基因植株相比,RAP2L15在转基因株系中的表达水平更高。对转基因株系和非转基因植株的株高测量发现,与非转基因植株相比,转基因植株的株高显著小于非转基因对照,表明RAP2L15的过量表达明显抑制了杨树株高生长。本研究将为进一步深入分析转录因子RAP2L15在调控杨树生长发育方面的功能提供参考依据与材料基础。Poplar propagation fast,easy cultivation,has been widely used in ecological shelterbelt and industrial timber forest.ERF(ethylene responsive factor)transcription factor is the largest family in AP2/ERF family.It plays an important role in regulating plant growth and development and responding to various abiotic stresses.The purpose of this paper is to understand the effect of transcription factor RAP2L15 on the regulation of poplar growth and development.In this study,'717 poplar'was used as material to clone the total cDNA sequence of ERF transcription factor RAP2L15,and the RAP2L15 overexpression vector was constructed.The results show that,fourteen transgenic lines were successfully obtained using Agrobacterium-mediated transformation of poplar.The RT-qPCR results showed that the expression level of RAP2L15 in transgenic strains was higher than that in non-transgenic plants.The plant height measurements of transgenic and non-transgenic plants showed that compared with non-transgenic plants,the plant height of transgenic plants was significantly lower than that of non-transgenic plants,indicating that overexpression of RAP2L15 significantly inhibited the growth of poplar plant height.This study will provide reference and material basis for further analysis of the function of transcription factor RAP2L15in regulating the growth and development of poplar.
