摘 要:水稻中的硝酸盐转运蛋白基因NRT1.1B可以调节水稻氮肥利用效率,影响水稻产量。为了提升NRT1.1B基因型鉴定和育种应用效率,本研究开发了一个可以高通量鉴定NRT1.1B基因型的分子标记NRT1.1B-KASP。利用NRT1.1B-KASP对籼稻‘9311’和粳稻‘繁14’的回交BC4F2群体进行NRT1.1B基因的分型,发现在BC4F2群体的65个单株的DNA样品中,有14份样品与供体亲本‘9311’同为NRT1.1B-indica基因型,17份样品与‘繁14’同为NRT1.1B-japonica基因型,34份样品显示杂合基因型。对这65份DNA样品的NRT1.1B基因进行测序,测序结果显示与KASP标记分型的结果完全一致。以上结果说明NRT1.1B-KASP标记可以高效、准确地对NRT1.1B进行基因分型,可以应用于水稻分子标记辅助选择育种。The nitrate transporter gene NRT1.1B in rice can regulate nitrogen-use efficiency and affect rice yield.In order to improve the efficiency of genotyping NRT1.1B and its breeding application,a molecular marker NRT1.1B-KASP which can identify NRT1.1B genotype high throughput was developed.NRT1.1B-KASP was used to genotype the NRT1.1B gene in a BC4F2population,derived from indica rice'9311'crossed with japonica rice'Fan14'.It was found that among the 65 individual DNA samples of BC4F2population,14 samples were homozygous for NRT1.1B-indica same as the donor parent'9311',17 samples were homozygous for NRT1.1Bjaponica same as the recurrent parents'Fan14',while 34 samples showed heterozygous genotype.The NRT1.1B gene of these 65 DNA samples was sequenced,and the sequencing results were completely consistent with the results of KASP marker genotyping.These results indicate that NRT1.1B-KASP marker can genotype NRT1.1B efficiently and accurately,so it can be applied to rice molecular marker assisted selection breeding.
