摘　　要：目的 :探讨葛根素调节肿瘤免疫微环境对卵巢癌Caov3细胞生长、增殖、凋亡、侵袭的影响。方法 :不同浓度葛根素处理THP-1细胞上清作用于Caov3细胞,CCK8实验检测Caov3细胞生长;ELISA检测上清液中白细胞介素(IL)-4、IL-10、IL-6的含量;RT-qPCR检测THP-1细胞中IL-4、IL-10、IL-6 mRNA的表达;克隆形成实验检测Caov3细胞克隆数量;流式细胞术检测Caov3细胞凋亡;免疫印迹检测Caov3细胞凋亡标记蛋白(cleaved caspase-3/caspase-3、cleavedcaspase-9/caspase-9)的表达;Transwell实验检测Caov3细胞侵袭;免疫印迹检测Caov3细胞侵袭相关蛋白血管内皮生长因子、纤维黏连蛋白、波形蛋白的表达。结果 :与0μmol/L相比,葛根素浓度升高至10μmol/L时,Caov3细胞活力受到显著抑制;与0μmol/L组相比,10、20μmol/L剂量组IL-6的含量及mRNA表达水平、凋亡率均显著升高,克隆细胞形成率、侵袭细胞数、侵袭相关蛋白表达均显著降低;IL-4、IL-10结果则相反。结论 :葛根素可影响肿瘤免疫微环境,抑制卵巢癌Caov3细胞生长、侵袭,并促进凋亡,缓解卵巢癌肿瘤的发生与发展。Objective:To investigate the effects of puerarin on the growth,proliferation,apoptosis and invasion of ovarian cancer Caov3 cells by regulating tumor immune microenvironment.Methods:Caov3 cells were treated with supernatants from THP-1 cells treated with different concentrations of puerarin.CCK8 assay was used to detect the growth of ovarian cancer cells.ELISA was utilized to detect the content of interleukin(IL)4,IL-10,and IL-6.RT-PCR was adopted to detect the mRNA expression of IL-4,IL-10,and IL-6.Colony formation assay was adopted to detect the number of cell colonies.Flow cytometry was applied to detect cell apoptosis.Western blotting was adopted detect the expression of apoptosis marker proteins(caspase-3,caspase-9).Transwell chamber was employed detect cell invasion.Western blotting was used to detect the expression of invasion-related proteins(vascular endothelial growth factor,fibronectin,vimentin).Results:Compared with the control group,when the puerarin concentration was 10μmol/L,the viability of ovarian cancer Caov3 cells was significantly inhibited.Compared with the 0μmol/L group,the levels and mRNA expression of IL-6,as well as the apoptosis rate,were significantly increased,while clone formation rate,number of invasion cells,and invasion-related protein expression were all significantly reduced.The results for IL-4 and IL-10 were opposite.Conclusion:Puerarin modulates tumor immune microenvironment to inhibit the growth and invasion of ovarian cancer Caov3 cells and to promote their apoptosis,thereby alleviating the occurrence and development of ovarian cancer tumors.
