摘 要:目的本研究旨在探讨不同栽培条件对铁皮石斛多糖含量的影响,并根据实验结果提出提升铁皮石斛多糖含量的栽培方法。方法本研究通过苯酚—硫酸法提取铁皮石斛中的多糖,并利用紫外可见分光光度法测定其吸光度,绘制标准曲线并计算多糖含量。实验样品分别来自不同栽培条件(大棚栽培与附树栽培)、不同海拔(500m、1000m、1500m)、同一栽培方式下不同部位(根、茎、叶)、不同栽培时间(6个月、12个月、18个月)的铁皮石斛,通过这些因素对多糖含量的影响进行对比分析。结果实验结果表明:一是大棚栽培铁皮石斛的多糖含量显著高于附树栽培;二是随着海拔的增加,铁皮石斛的多糖含量逐步增加;三是茎部是铁皮石斛中多糖含量的主要积累部位;四是在相同部位下,大棚栽培的多糖含量普遍高于附树栽培;五是随着栽培时间的延长,铁皮石斛的多糖含量逐渐增加。结论本研究表明,栽培条件、海拔、部位选择及栽培时间等因素均对铁皮石斛的多糖含量有显著影响。为了提高铁皮石斛的多糖含量,应选择大棚栽培模式,适宜的海拔(如1000m以上),重点关注茎部的养护,适当延长栽培周期。上述方法为铁皮石斛的栽培管理及其药用成分的优化提供了理论依据,具有重要的应用价值。Objective This study aims to explore the effects of different cultivation conditions on the polysaccharide content of Dendrobium officinale,and propose cultivation methods to increase the polysaccharide content of Dendrobium officinale based on experimental results.Method This study extracted polysaccharides from Dendrobium officinale using the phenol sulfuric acid method,and measured their absorbance using UV visible spectrophotometry.A standard curve was drawn and the polysaccharide content was calculated.The experimental samples were collected from Dendrobium officinale under different cultivation conditions(greenhouse cultivation and attached tree cultivation),different altitudes(500m,1000m,1500m),different parts(roots,stems,leaves)under the same cultivation method,and different cultivation times(6 months,12 months,18 months).The effects of these factors on polysaccharide content were compared and analyzed.Result The experimental results showed that:firstly,the polysaccharide content of Dendrobium officinale grown in greenhouses was significantly higher than that grown in attached trees;Secondly,as the altitude increases,the polysaccharide content of Dendrobium officinale gradually increases;Thirdly,the stem is the main accumulation site of polysaccharide content in Dendrobium officinale;Fourthly,under the same location,the polysaccharide content in greenhouse cultivation is generally higher than that in attached tree cultivation;With the extension of cultivation time,the polysaccharide content of Dendrobium officinale gradually increases.Conclusion This study indicates that factors such as cultivation conditions,altitude,site selection,and cultivation time have a significant impact on the polysaccharide content of Dendrobium officinale.In order to increase the polysaccharide content of Dendrobium officinale,the greenhouse cultivation mode should be selected,with a suitable altitude(such as over 1000m),focusing on stem maintenance,and appropriately extending the cultivation period.The above methods provide th
