色差法联合显微成像技术研究枸杞颜色与其药效成分的相关性

摘  要:本研究旨在探究枸杞颜色、显微特征颜色与其药效成分之间的相关性。借助显微成像技术联合色差法分别收集30批次枸杞样品显微特征颜色及其颜色信息,采用紫外分光光度法和高效液相色谱法检测各样品中枸杞多糖、甜菜碱、绿原酸、阿魏酸、槲皮苷含量,联合Pearson法分析枸杞颜色、显微特征颜色与其各药效成分间相关性。枸杞外果皮表皮细胞L^(*)值与枸杞颜色L^(*)、a^(*)、b^(*)、E_(ab)^(*)值,呈现显著低度负相关(P<0.05);中果皮薄壁细胞L^(*)值与枸杞颜色L^(*)、a^(*)、b^(*)、E_(ab)^(*)值,呈现低度负相关,且与b^(*)、E_(ab)^(*)值存在显著性(P<0.05);中果皮薄壁细胞b^(*)值与枸杞颜色L^(*)、a^(*)、b^(*)、E_(ab)^(*)值,呈现低度正相关,且与a^(*)、b^(*)、E_(ab)^(*)值存在显著性(P<0.05);中果皮薄壁细胞E_(ab)^(*)值与枸杞颜色a^(*)、b^(*)、E_(ab)^(*)值,呈现低度负相关,且与a^(*)、b^(*)值存在显著性(P<0.05)。绿原酸含量与枸杞a^(*)、E_(ab)^(*)值呈低度负相关;阿魏酸含量与枸杞a^(*)、E_(ab)^(*)值呈低度正相关,且与a^(*)值存在显著性(P<0.05);槲皮苷含量与枸杞L^(*)值呈低度负相关。枸杞多糖含量与外果皮表皮细胞a^(*)值呈低度正相关;甜菜碱含量与种皮石细胞b^(*)值呈低度负相关;绿原酸含量与中果皮薄壁细胞b^(*)值呈低度负相关;阿魏酸含量与外果皮表皮细胞L^(*)、E_(ab)^(*)值呈低度负相关,与外果皮表皮细胞b^(*)值呈低度正相关,且与L^(*)、b^(*)值存在极显著性(P<0.01)。枸杞颜色及显微特征颜色与枸杞药效成分含量具有一定相关性,可为药材质量综合评价提供支撑。This study aims to investigate the correlation between the external color,microscopic characteristic color and the pharmacodynamic component content of Lycii Fructus.Based on 30 batches of Lycii Fructus,the external color value was determined by color difference method,the microscopic characteristic color information was determined by microscopic imaging technology,the pharmacodynamic component content was determined by ultraviolet-visible spectrophotometry method,and HPLC method.Pearson correlation analysis was used to analyze the correlation between external color,microscopic characteristic color and the pharmacodynamic component content of Lycii Fructus.There was a significant negative correlation between the L^(*)value of epidermal cells in outer pericarp and the L^(*),a^(*),b^(*),E_(ab)^(*) values of color of Lycii Fructus(P<0.05).The values of L^(*)in mesocarp parenchyma cells were negatively correlated with the values of L^(*),a^(*),b^(*),E_(ab)^(*),and were significantly correlated with the values of b^(*),E_(ab)^(*)(P<0.05).The b^(*)value of mesocarp parenchyma cell was positively correlated with L^(*),a^(*),b^(*),E_(ab)^(*) values,and significantly correlated with a^(*),b^(*),E_(ab)^(*) values(P<0.05).The E_(ab)^(*) values of mesocarp parenchyma cells were negatively correlated with the a^(*),b^(*)and E_(ab)^(*) values of the color of Lycii Fructus,and were significantly correlated with the a^(*)and b^(*)values(P<0.05).Chlorogenic acid content was negatively correlated with a^(*)and E_(ab)^(*) values of Lycii Fructus.Ferulic acid content was positively correlated with a^(*)and E_(ab)^(*) values,and significantly correlated with a^(*)values(P<0.05).There was a low negative correlation between quercetin content and L^(*)value of Lycii Fructus.The content of Lycii Fructus polysaccharide was positively correlated with the a^(*)value of exocarp epidermal cells.The content of betaine was negatively correlated with the b^(*)value of seed stone cells.Chlorogenic acid content was negatively correlated with b^(*)
