摘  要:[目的]利用网络药理学方法,研究茶树精油治疗鸡肝损伤的作用机制。[方法]通过TCMSP数据库检索筛选出已检测出成分的作用靶点;通过GeneCard、DisgeNET和OMIM数据库查询鸡肝损伤的相关靶点信息;使用Cytoscape软件构建成分-靶点和茶树精油-成分-靶点-鸡肝损伤图;绘制药物靶点和疾病靶点韦恩图,构建交集靶点互作网络图,随后对交集靶点进行GO和KEGG富集分析。利用String数据库,筛选核心靶点并用AutoDock软件进行分子对接。[结果]TCMSP数据库筛选出4个有效活性成分,对应60个靶点基因。对不同疾病数据库进行搜索,共获得1 656个鸡肝损伤疾病靶点。韦恩图结果显示药物和疾病交集靶点共28个,通过界值筛选得到11个核心靶点,其中3个为关键核心靶点,主要为CYP19A1、ESR1、ESR2P53等。进行GO和KEGG富集分析,KEGG结果显示靶点主要涉及的信号通路包括PPAR信号通路、化学致癌-受体激活、雌激素信号通路等。[Objective]To investigate the mechanism of tea tree essential oil in treating chicken liver injury using network pharmacology methods.[Method]Retrieve and screen the action targets of the detected components through the TCMSP database;Query the relevant target information of chicken liver injury through GeneCard,DisgeNET,and OMIM databases;Construct component target and tea tree essential oil component target chicken liver injury maps using Cytoscape software;Draw a Venn diagram of drug targets and disease targets,construct an intersection target interaction network diagram,and then perform GO and KEGG enrichment analysis on the intersection targets.Using a String database,screen core targets and perform molecular docking using AutoDock software.[Results]Four effective active ingredients were screened from the TCMSP database,corresponding to 60 target genes.A total of 1656 chicken liver injury disease targets were obtained through searching different disease databases.The Venn diagram results showed a total of 28 intersecting targets between drugs and diseases,and 11 core targets were identified through threshold screening.Among them,3 were key core targets,mainly CYP19A1,ESR1,ESR2P53,etc.GO and KEGG enrichment analysis were conducted,and KEGG results showed that the target mainly involves signaling pathways such as PPAR signaling pathway,chemo oncogenic receptor activation,estrogen signaling pathway,etc.
