摘 要:本研究旨在建立膨化宠物食品中姜黄素、去甲氧基姜黄素、双去甲氧基姜黄素含量的高效液相色谱检测方法。使用C18色谱柱(250 mm×4.6 mm×5μm),流动相为乙腈:甲酸(0.1%)=65:35,提取剂为甲醇,流速为1.0 mL/min,柱温为30℃,检测波长为425 nm,进样量为10μL。检测姜黄素类化合物使用甲醇超声溶解30 min,过膜后上机检测。结果显示,该方法检测姜黄素、去甲氧基姜黄素、双去甲氧基姜黄素分别在4.24~84.83μg/mL、1.70~68.00μg/mL、5.64~84.56μg/mL范围内线性良好,相关系数分别为0.9999、0.9991、0.9997。检出限分别为0.020μg/mL、0.013μg/mL、0.017μg/mL,定量限分别为0.080μg/mL、0.0544μg/mL、0.068μg/mL。该方法操作简单、成本低、准确度高、精密度良好,适用于膨化宠物食品中姜黄素类化合物含量的测定。An efficient high-performance liquid chromatography(HPLC)method was established for the determination of curcumin,demethoxycurcumin,and bisdemethoxycurcumin in dog feed.The HPLC was performed using a C18 column(250 mm×4.6 mm×5μm)with a mobile phase consisting of acetonitrile and 0.1%formic acid in a ratio of 65:35.Methanol was used as the extraction solvent,with a flow rate of 1.0 mL/min,a column temperature of 30°C,and a detection wavelength of 425 nm.A sample volume of 10μL was injected.The curcumin compounds were dissolved in methanol using ultrasonic extraction for 30 minutes,followed by filtration before analysis.The results indicated that the method exhibited good linearity for curcumin,demethoxycurcumin,and bisdemethoxycurcumin in the concentration ranges of 4.24~84.83μg/mL,1.70~68.00μg/mL,and 5.64~84.56μg/mL,respectively.The limits of detection were 0.020μg/mL,0.013μg/mL,and 0.017μg/mL,while the quantification limits were 0.080μg/mL,0.0544μg/mL,and 0.068μg/mL,respectively.This method is easy-operation,cost-efficient,highly accurate,and with excellent precision,which is proper for the determination of curcumin compounds in dog feed.
