摘　　要：目的:考察自制乌头茎叶洗液的抗炎镇痛作用及其皮肤给药安全性,以期为该制剂进一步开发为用于类风湿性关节炎的外用洗液提供药理学依据。方法:采用热板法评价自制洗液对小鼠的镇痛作用;采用角叉菜胶致小鼠足肿胀试验,并使用ELISA法检测小鼠足肿胀组织中TNF-α、IL-6和IL-1β水平以评价其抗炎作用;通过新西兰兔局部给药刺激性试验评价其刺激性;用健康小鼠评价其皮肤急性毒性。结果:热板法致痛试验中,高剂量的药液能延长小鼠的痛阈值(P<0.01)。抗炎试验中,中、高剂量药液可以抑制造模后足肿胀程度(P<0.05);高、中剂量药液可以降低足肿胀组织中TNF-α、IL-6和IL-1β水平(P<0.05)。病理切片显示,随着给药剂量的升高,中性粒细胞逐渐减少。皮肤刺激性试验预试结果暂未显示洗液对新西兰兔有皮肤刺激性,有待后续进一步研究。皮肤急性毒性试验结果显示,洗液对小鼠皮肤无毒性。结论:自制的乌头茎叶洗液具有明显的抗炎镇痛作用,且外用安全性较高,有进一步开发为用于类风湿性关节炎外用洗液的价值。Objective:To investigate the anti-inflammatory and analgesic effects of a self-prepared Aconitum carmichaelii Debx.stem and leaf lotion,and to evaluate its skin administration safety,with the aim of providing a pharmacologic basis for further development of the preparation into a topical lotion for rheumatoid arthritis.Methods:The analgesic effect of self-made lotion on mice was assessed using the hot plate method;the anti-inflammatory effect was evaluated using the carrageenan-induced paw edema model in mice,with employed to measure the levels of TNF-α,IL-6,and IL-1βin the paw edema tissue.The skin irritation was assessed through local application in New Zealand rabbits,and the acute dermal toxicity was evaluated in healthy mice.Results:In the pain-inducing experiment using the hot plate method,the high-dose lotion significantly prolonged the pain threshold in mice.In the anti-inflammatory experiment,both medium and high doses of the lotion inhibited the degree of paw edema after administration;the high-dose lotion significantly reduced the levels of TNF-α,IL-6 and IL-1βin the paw edema tissues,while the medium dose significantly reduced the levels of TNF-αand IL-6.Pathological sections showed a gradual decrease in neutrophils with increasing doses.The pre-test results of skin irritation experiment did not show that the lotion had skin irritation on New Zealand rabbits,which needs to be further studied.The results of acute skin toxicity test showed that the lotion had no toxicity to the skin of mice.Conclusion:The self-prepared Aconitum carmichaelii Debx.stem and leaf lotion exhibits significant anti-inflammatory and analgesic effects,with high safety for topical use.It has the value of further development as a topical lotion for rheumatoid arthritis.
