摘  要:目的:以植物来源的玉米醇溶蛋白为载体材料制备黄芩苷-玉米醇溶蛋白纳米粒(baicalin-zein nanoparticles,BAZNPs),并对其进行体外抗肿瘤活性评价。方法:采用相分离法构建BA-ZNPs,通过观测形态学、粒径大小、Zeta电位、包封率、载药量等指标对纳米粒进行表征。通过CCK-8法检测BA-ZNPs对转移性人乳腺癌细胞MDA-MB-231生长情况的影响;细胞划痕实验和Transwell小室评价BA-ZNPs对细胞迁移和侵袭作用的影响;流式细胞仪检测BA-ZNPs细胞周期分布及细胞凋亡情况。结果:BA-ZNPs呈类球形,粒径约为110 nm,包封率为(83.02±1.24)%,载药量为(4.39±0.32)%。载体材料无细胞毒性,黄芩苷(baicalin,BA)、BA-ZNPs均可显著抑制肿瘤细胞的生长,纳米化显著增强BA的细胞毒作用。相较于BA,BA-ZNPs对肿瘤细胞愈合能力的抑制作用显著增强,对肿瘤细胞的迁移和侵袭抑制作用也显著提高,更利于发挥细胞周期阻滞作用,提高BA对肿瘤细胞的杀伤能力并诱导细胞凋亡。结论:黄芩苷纳米粒可有效抑制MDA-MB-231细胞的增殖、迁移和侵袭,进而诱导细胞凋亡,有望为治疗转移性乳腺癌提供基础理论依据。OBJECTIVE To prepare baicalin-zein nanoparticles(BA-ZNPs)using plant-derived zein as a carrier material and to evaluate their in vitro anti-tumor activity.METHODS BA-ZNPs were synthesized using the phase separation method,and characterized by morphological observation,particle size,zeta potential,encapsulation efficiency and drug loading.The effect of BA-ZNPs on the proliferation of the metastatic human breast cancer cell line MDA-MB-231 was detected by cell counting kit-8(CCK-8)assay.Cell scratch assay and Transwell assay were used to evaluate the effects of BA-ZNPs on cell migration and invasion,respectively.Flow cytometry was employed to investigate the cell cycle distribution and apoptosis induced by BA-ZNPs.RESULTS BA-ZNPs were spherical in shape,with an average particle size of approximately 110 nm,an encapsulation efficiency of(83.02±1.24)%,and a drug loading capacity of(4.39±0.32)%.The carrier material was non-cytotoxic.Both BA and BA-ZNPs displayed significant inhibitory effects on tumor cell growth.Nanoization significantly enhanced the cytotoxicity of BA.Compared to BA,BA-ZNPs had a significantly stronger inhibitory effect on the healing ability,migration,invasion and cell cycle arrest of tumor cells,which also enhanced the kill effect on tumor cells and induced cell apoptosis.CONCLUSION BAZNPs can effectively inhibit the proliferation,migration,and invasion of MDA-MB-231 cells,and then induce apoptosis.This study is expected to provide a theoretical basis for the treatment of metastatic breast cancer.
