摘  要:采用高温-菌丝尖端处理技术对1株退化的灵芝菌株进行纯化和复壮,通过菌丝生长情况、菌丝的显微形态、菌丝中漆酶和羧甲基纤维素酶活性、栽培后子实体的农艺性状等指标评价复壮效果。结果表明,复壮后菌株生长情况良好,菌丝无色透明、有横膈膜、粗细不均、锁状联合明显、尖端呈放射性分支,菌管表面光滑无纹饰、管间清晰无杂质。复壮菌株菌丝生长速度是原代菌株的1.46倍;与原代菌株相比漆酶活性提高了60%,羧甲基纤维素酶活性提高了85%。栽培试验表明复壮菌株子实体的农艺性状优于原代菌株,菌包污染率下降了25%,生物学效率提高了3.38%。Purification and rejuvenation of a degenerating strain of Ganoderma lingzhi was carried out by the technique of high temperature and hyphal tip treatment.The effect of rejuvenation was evaluated by the growth of mycelia,the microscopic morphology of mycelia,the activities of laccase and carboxymethyl cellulase in mycelia,and the agronomic traits of fruit bodies after cultivation.The results showed that the strain grew well after rejuvenation,the hyphal was colorless and transparent,with diaphragms,uneven thickness,obvious clamp connection,radioactive branches at the tip,the surface of the tube is smooth and no decoration,and the intertube was clear and inclusion-free.The mycelial growth rate of the rejuvenated strain was 1.46 times that of the original strain.The enzymes activities of laccase and carboxymethyl cellulase increased by 60%and 85%respectively.Cultivation test shows that all the agronomic characters of fruit bodies were better than those of the original strain,and pollution rate decreased by 25%,biological efficiency increased by 3.38%.
