摘 要:“玉农76”玉米杂交种是河南农业职业学院选育并大面积推广种植的玉米品种。研究利用40对玉米SSR核心引物对2个待测样品进行DNA指纹鉴定,确定高产、优质玉米品种“玉农76”的真实性,发现SSR标记umc2007y4、bnlg1940k7、umc1705w1、bnlg2305k4、bnlg1702k1、bnlg240k1、phi080k15、umc1432y6、bnlg1520k1、umc1999y3、umc2160k3、bnlg2235y5、umc2084w2、umc2163w3能够在待测样品和“玉农76”间扩增出清晰、稳定的条带,认为这14对SSR引物可用于“玉农76”品种真实性的快速鉴定;发现4对共显性SSR标记phi053k2、bnlg2291k4、umc1125y3、bnlg1520k1能够在“玉农76”及其亲本自交系间扩增出清晰、稳定的共显性条带,认为这4对SSR引物可用于“玉农76”的种子纯度鉴定;发现标记bnlg1520k1可同时对“玉农76”的品种真实性和种子纯度进行鉴定。此外,研究构建了“玉农76”及其亲本自交系的十进制数字指纹图谱,为加强“玉农76”的品种保护及种子品质监管提供了理论依据与技术支持。"Yunong 76"is a major maize hybrid variety,which was selected and cultivated by Henan Vocational College of Agriculture.To determine the authenticity of the high yield and quality maize variety"Yunong 76",two test varieties were assessed by DNA fingerprinting using 40 pairs of maize SSR core primers.The result showed that 14 pairs of SSR primers(umc2007y4,bnlg1940k7,umc1705w1,bnlg2305k4,bnlg1702k1,bnlg240k1,phi080k15,umc1432y6,bnlg1520k1,umc1999y3 and umc2160k3)could amplify clear and stable bands between the two-test sample and"Yunong 76".Therefore,these 14 pairs of SSR primers were able for rapid authenticity assessment of"Yunong 76".Meanwhile,4 pairs of co-dominant SSR primers(phi053k2,bnlg2291k4,umc1125y3,bnlg1520k1)could amplify clear and stable co-dominant bands between"Yunong 76"and its parent inbred lines.Thus,these 4 pairs of SSR primers can be used for purity identification of"Yunong 76".The SSR primer bnlg1520k1 can be used to identify both authenticity and purity of"Yunong 76".This study also constructed a decimal digital fingerprint maps of"Yunong 76"and its parents,providing the theo⁃retical basis and technical support for strengthening the variety protection of"Yunong 76"and seed quality supervision.
