摘 要:为探索海水替代淡水以达到节约淡水资源培养微藻的可行性,本研究将人工海水稀释2、3、6倍后再补加氮、磷和铁盐,用来培养魏氏真眼点藻Vischeria sp. WL1.研究发现:在所有稀释后的培养基中,稀释2倍海水培养的Vischeria sp. WL1具有最大产油量;人工海水培养下该藻积累的油脂中以C16∶1(60%)和C18∶1(16%)脂肪酸为主,C20∶5(EPA)仅占5.00%.通过向稀释2倍海水中补加甘油、乙酸钠、乙酸铵三种碳源,结果表明:补加4 mM乙酸铵是促进Vischeria sp. WL1生长和产油的最佳条件,油脂中C16∶0和C16∶1脂肪酸比例最高;补加8 mM乙酸钠条件下所获油脂具有最高的EPA含量,可达9.72%.因此,通过人工海水来代替淡水资源并添加必需营养元素和合适的碳源,是培养产油微藻的一条可选途径.To explore the feasibility of using seawater to replace fresh water for saving fresh water resources and cultivating microalgae,in this study,artificial seawater was diluted by 2,3,and 6 times respectively,and then nitrogen,phosphorus,and iron salts were added to culture Vischeria sp.WL1.The study found that among all the diluted media,Vischeria sp.WL1 cultured in seawater diluted by two times had the largest oil production.Under artificial seawater cultivation,the oil accumulated by this alga is mainly composed of C16∶1(60%)and C18∶1(16%)fatty acids,and C20∶5(EPA)only accounts for 5.00%.By adding three carbon sources of glycerol,sodium acetate,and ammonium acetate to the seawater diluted by two times,the results show that adding 4 mM ammonium acetate is the best condition for promoting the growth and oil production of Vischeria sp.WL1,and the proportion of C16∶0 and C16∶1 fatty acids in the oil is the highest.Under the condition of adding 8 mM sodium acetate,the obtained oil has the highest EPA content,reaching up to 9.72%.Therefore,using artificial seawater to replace fresh water resources and adding necessary nutrients and appropriate carbon sources is an alternative way to cultivate oil-producing microalgae.
