摘　　要：为探究环状RNA CircFoxO3的表达对猪成肌细胞增殖和凋亡的影响,本试验利用CircFoxO3过表达载体及siRNA转染猪成肌细胞,通过EdU和Annexin V-mCherry检测成肌细胞增殖和凋亡情况,利用RT-qPCR检测凋亡相关基因的表达。结果表明,猪成肌细胞过表达CircFoxO3后增殖细胞数量减少,凋亡细胞数量上升,凋亡相关基因Caspase-3、Caspase-8、Fas、FasL表达量显著上调(P<0.05),Bax表达量极显著上调(P<0.01),抗凋亡基因Bcl-2表达量显著下调(P<0.05);转染CircFoxO3siRNA后,成肌细胞凋亡水平下降,增殖能力增加,Bax、FasL基因表达量极显著下调(P<0.01),Caspase-3、Caspase-8表达量显著降低(P<0.05),Bcl-2表达显著增加(P<0.05)。说明Circ-FoxO3表达抑制了猪成肌细胞的增殖,促进了凋亡相关基因的表达,从而促进成肌细胞的凋亡。In order to explore whether the expression of CircFoxO3is related to the proliferation and apoptosis of porcine myoblasts,this study transfected porcine myoblasts with CircFoxO3overexpression vector and siRNA,and detected cell proliferation and apoptosis by EdU and Annexin VmCherry fluorescence staining,and detected the expression of apoptosis-related genes by RTqPCR.The results showed that after transfection with overexpression vector,the proportion of apoptotic cells increased and the number of cells proliferation decreased.At the same time,the expression of apoptosis-related genes Caspase-3,Caspase-8,Fas and FasL was significantly up-regulated by RT-qPCR(P<0.05),and the expression of Bax was extremely significantly up-regulated(P<0.01).The expression of Bcl-2was significantly down-regulated(P<0.05).After transfection with si-CircFoxO3,the apoptosis level decreased and proliferation ability increased.the expression of Bax and FasL genes was highly significantly down-regulated(P<0.01),the expression of caspase-3and Caspase-8was significantly decreased(P<0.05),and the expression of Bcl-2was significantly increased(P<0.05).CircFoxO3expression inhibited proliferation of porcine myoblasts,promoted the expression of apoptosis-related genes,and promoted apoptosis of myoblasts.
