摘  要:为了探究白术多糖(polysacchcride of Atractylodes macrocephala koidz,PAMK)对脂多糖(LPS)诱导的蛋雏鸡急性肾损伤的缓解作用。将100只1日龄雄性海兰褐蛋鸡随机分为5组,即空白对照组(CON组)、LPS处理组(LPS组)、PAMK低剂量组(LPS+PAMK 250mg/L,A组)、PAMK中剂量组(LPS+PAMK 500mg/L,B组)和PAMK高剂量组(LPS+PAMK 1000mg/L,C组)。1~20日龄,PAMK处理组每天混水饲喂相应剂量PAMK。在第14、16、18、20日龄时,LPS组与PAMK处理组鸡只腹腔注射LPS,空白对照组注射等体积生理盐水。饲养至21日龄采集肾脏,HE染色法观察肾脏组织切片病理变化;测量肾质量以及雏鸡体质量计算肾脏系数;测定肾脏组织中尿素氮(BUN)、肌酐(CRE)、丙二醛(MDA)、超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)和总抗氧化能力(TAOC)含量;ELISA法测定TNF-α和IL-1β含量;实时荧光定量PCR技术检测肾脏中炎症因子基因IL-1β、TNF-α和NF-κB的mRNA表达量,及肾脏中细胞凋亡因子p53、Bcl-2、Bax和Caspase-3的mRNA表达量。结果表明:显微结构观察结果显示,PAMK缓解了LPS引起的肾脏组织损伤;与LPS组相比,PAMK可显著降低肾脏系数和肾脏BUN浓度(P<0.05);与LPS组相比,PAMK组的TNF-α、IL-1β含量以及TNF-α、IL-1β、NF-κB的mRNA表达量均降低;与LPS组相比,PAMK组T-AOC、SOD活力和GSH-Px活性显著增加(P<0.05),MDA含量极显著降低(P<0.01);与LPS组相比,PAMK低、中剂量组的Bax、Caspase-3和p53的mRNA表达量降低,Bcl-2的mRNA表达量升高。综上所述,PAMK可抑制LPS造成的炎性反应和氧化应激反应,缓解肾脏细胞的凋亡,且PAMK中剂量组治疗效果最好,为PAMK治疗鸡急性肾损伤提供了理论依据。To investigate the effect of the polysaccharide derived from Atractylodes macrocephala koidz.(PAMK)on lipopolysaccharide(LPS)-induced acute kidney injury in juvenile laying hens.A total of 100one-day-old male Hy-Line brown laying hens were randomly divided into five groups:a control group(CON group),an LPS treatment group(LPS group),a PAMK low-dose group(LPS+PAMK 250mg/L,group A),a PAMK medium-dose group(LPS+PAMK 500mg/L,group B),and a PAMK high-dose group(LPS+PAMK 1000mg/L,group C).The PAMK-treated groups were fed with corresponding doses of PAMK mixed with water daily from 1-20days of age.At the age of 14,16,18and 20days,the chickens in the LPS group and PAMK-treated groups were intraperitoneally injected with LPS,while the chickens in the blank control group were injected with the same volume of normal saline.After feeding for 21days,the kidneys were collected,and pathological changes in kidney tissue sections were observed using hematoxylin and eosin(HE)staining.Kidney weight and chick weight were measured to calculate the kidney coefficient.The levels of BUN,CRE,MDA,SOD,GSH-Px and T-AOC in kidney tissues were measured.The contents of TNF-αand IL-1βwere determined by ELISA.The mRNA expression of inflammatory factor genes(IL-1β,TNF-αand NF-κB)and apoptosis-related factors(p53,Bcl-2,Bax and Caspase-3)in the kidneys was detected using quantitative real-time PCR.The results indicated that PAMK alleviated LPS-induced renal tissue injury.Compared to the LPS group,PAMK significantly reduced the kidney coefficient and BUN concentration in the kidneys(P<0.05).Additionally,the levels of TNF-αand IL-1β,as well as the mRNA expression of TNF-α,IL-1β,and NF-κB in the PAMK group,were decreased compared to the LPS group.Furthermore,T-AOC,SOD and GSHPx activities in the PAMK group were significantly increased(P<0.05),while MDA content was significantly decreased(P<0.01).Compared with the LPS group,the mRNA expression of Bax,Caspase-3and p53in the PAMK low-dose group and the PAMK medium-dose group decreased,while the
