摘 要:禾本科(Gramineae)薏苡属(Coix)植物薏苡(Coix lacryma-jobi L.)为著名的药食两用农作物,前期主要开展了薏苡果实的药学研究,关于其根茎等其他部位的化学成分发现较少报道,本研究综合利用硅胶、ODS、MCI、HPLC色谱等分离手段,对薏苡根水提物正丁醇部位进行了分离纯化,并利用NMR以及MS等现代波谱学方法对结构进行了确定。最终从薏苡根中分离到7个单体化合物,分别为methyl-3-(4-hydroxyphenyl) propionate(1),dihydroferulic acid methyl ester(2),3-β-D-glucopyranosyl-6-methoxy-2-benzoxazolinone (3),N-trans-feruloyl tyramin (4),trans-N-p-coumaroyl tyramine (5),(2R)-2-O-β-D-glucopyranosyl-2H-1,4-benzoxazin-3(4H)-one(6),(2S)-2-O-β-D-glucopyranosyl-2H-1,4-benzoxazin-3(4H)-one(7)。其中,化合物4在100μmol/L浓度下具有较好的抗氧化活性,其DPPH自由基清除率为75.80±3.98%。Coix lacryma-jobi L.(family Gramineae, genus Coix) is a well-known medicinal and edible crop. Previous research has focused largely on the seeds of Coix, with relatively few reports on the chemical components of its roots and other parts. In this study, the n-butanol fraction of a water extract from Coix roots was separated and purified using silica gel, ODS, MCI, and HPLC chromatographic techniques. The structures of the isolated compounds were elucidated by modern spectroscopic methods, including NMR and MS. As a result, seven compounds were isolated and identified from Coix roots: methyl-3-(4-hydroxyphenyl) propionate(1), dihydroferulic acid methyl ester(2), N-trans-sinapoyltyramine(3), trans-N-p-coumaroyl tyramine(4), 3-β-D-Glucopyranosyl-6-methoxy-2-benzoxazolinone(5),(2R)-2-O-β-D-glucopyranosyl-2H-1,4-benzoxazin-3(4H)-one(6),(2S)-2-O-β-D-glucopyranosyl-2H-1,4-benzoxazin-3(4H)-one(7). Among these, compound 4 showed marked antioxidant activity at a concentration of 100 μmol/L, with a DPPH free radical scavenging rate of 75.80 ± 3.98%.
