摘 要:TIFY家族蛋白是一类在植物中广泛存在的转录因子,该家族蛋白含有一个高度保守的TIFY结构域,通过参与植物内部的多种信号途径,对植物的生长发育、逆境响应以及防御机制进行调节。本研究从‘草原4号’紫花苜蓿(MedicagosativaL.)中克隆得到MsTIFY77基因并对其进行生物学信息分析以及表达模式分析,探究MsTIFY77基因在胁迫应答中的功能。结果发现,该基因全长1101 bp,开放阅读框(Open reading frame,ORF)编码367个氨基酸,含有TIFY和CCT-2结构域。氨基酸序列分析表明MsTIFY77蛋白为不稳定蛋白,通过系统发育树分析表明紫花苜蓿MsTIFY77蛋白与蒺藜苜蓿(Medicago truncatula),豌豆(Pisum sativum),红三叶(Trifolium pratense),毛苕子(Vicia villosa),鹰嘴豆(Cicer arietinum)和白车轴草(Trifolium repens)的TIFY蛋白亲缘关系较近。亚细胞预测MsTIFY77蛋白定位于细胞核。qRT-PCR结果显示,低温和干旱胁迫诱导该基因的表达,而脱落酸胁迫对该基因表达有抑制作用。研究结果为MsTIFY77基因的功能提供参考。The TIFY family proteins are a class of transcription factors that widely presented in plants,which contain a highly conserved TIFY domain.They regulate plants growth and development,stress responses,and defense mechanisms by participating in numerous internal signaling pathways of plants.In this study,MsTIFY77 gene was cloned from Medicago sativa.L.‘Caoyuan No.4’and its expression pattern and biologi⁃cal information were preliminarily elucidated.The results showed that the gene is 1101 bp in length,with an open reading frame(ORF)encoding 367 amino acids,containing TIFY and CCT-2 domains.Amino acid sequence analysis showed that MsTIFY77 protein is an unstable protein.Phylogenetic tree analysis showed that MsTIFY77 protein is closely related to the TIFY proteins of Medicago truncatula,Pisum sativum,Trifo⁃lium pratense,Vicia villosa,Cicer arietinum and Trifolium repens.Subcellular localization indicated that the MsTIFY77 protein is located in the nucleus.The qRT-PCR results showed that low temperature and drought stress induced the expression of MsTIFY77 gene,while abscisic acid treatment inhibited its expression.The results provided a reference for the function study of MsTIFY77 gene.
