摘 要:核酸感受器在先天性免疫、炎症和肿瘤中具有重要的调控作用。DEAD-box RNA解旋酶(DEAD-box helicase)是哺乳动物基因中最大的RNA解旋酶家族,除具有解旋酶功能外,部分解旋酶分子也具有核酸感受器功能,可以对内源性或外源性异常核酸物质做出反应。对核酸感受器的认识也从维持正常生理功能到病原防御再上升到参与疾病的发生发展。维甲酸诱导基因Ⅰ样受体(Retinoic acid inducible gene-Ⅰreceptors,RLRs)主要包括维甲酸诱导基因I(Retinoic acid inducible gene-Ⅰ,RIG-Ⅰ)和黑色素瘤分化相关基因5(Melanoma differentiationassociated gene 5,MDA5),是解旋酶家族中被鉴定为参与RNA配体识别的核酸感受器,它们在病原防御、炎症、肿瘤中的作用被广泛研究。DExD/H-box解旋酶60(DExD/H-box helicase 60,DDX60)是另一个被鉴定为功能区域类似RIG-Ⅰ样受体的解旋酶分子,近年来其在肿瘤、炎症、病原防御中的作用及机制也逐渐被揭示,本文就DDX60的相关研究进展进行总结分析。Nucleic acid receptors have important regulatory roles in innate immunity,inflammation and tumors.The nucleic acid deconjugating enzymes DEAD-box RNA family is the largest family of RNA helicases in mammalian genes.In addition to their deconjugating enzyme functions,some of the deconjugating enzyme molecules also function as nucleic acid receptors that respond to endogenous or exogenous aberrant nucleic acid substances.The understanding of nucleic acid-sensing receptors has also advanced from the maintenance of normal physiological functions to pathogenic defense and to involvement in disease development.Retinoic acid inducible gene-Ⅰ(RIG-Ⅰ)-like receptors,mainly including RIG-Ⅰand melanoma differentiation-associated gene 5(MDA5),are among the family of deconjugating enzymes identified as nucleic acid receptors involved in the recognition of RNA ligands,and their roles in pathogenic defense,inflammation,and tumors have been extensively investigated.DExD/H-box helicase 60(DDX60)is another molecule that has been identified as having functional regions similar to those of the deconjugating enzyme molecule of the RIG-Ⅰ-like receptor.In recent years,its role and mechanism in tumor,inflammation,and pathogenic defense have been gradually revealed.This paper summarizes and analyses the progress of the research related to DDX60.
