罗布麻浸膏中黄酮类成分分析及其在混合型卷烟中的应用

摘　　要：[目的]研究罗布麻浸膏中6种黄酮类成分的定量分析方法,考察罗布麻浸膏主要功能成分转移行为以及对卷烟感官品质的影响和降低自由基的作用。[方法]以20 mL 70%甲醇-水为溶剂,ACQUITY UPLC HSS C_(18)(2.1 mm×100 mm,1.8μm)为分离柱,槲皮素-D5为内标,采用超声提取-UPLC-MS/MS方法,测定罗布麻浸膏中6种黄酮类成分含量。通过不同加料比例研究罗布麻浸膏对卷烟感官质量、减害作用的影响以及主要功能成分的转移行为。[结果]UPLC-MS/MS方法可在10 min内实现6种黄酮类成分的快速分离,标准工作曲线线性关系较好(R^(2)>0.9990),方法的LOD和LOQ分别为0.39~0.76和1.29~2.55 ng/mL,加标回收率为93.11%~105.77%,RSD为0.73%~2.89%。添加罗布麻浸膏可以提升卷烟香气质量,降低烟气刺激性;主流烟气中槲皮素转移率为0.89%~2.93%,气相自由基清除率最大可达30%。[结论]该方法操作简便、响应快速、准确度高、重复性好,可适用于罗布麻浸膏中活性成分的快速测定。卷烟加料应用结果表明,罗布麻浸膏可以有效抑制卷烟主流烟气中自由基的释放量,合适的施加比例(≤1.0%)可以有效提升混合型卷烟感官品质并保持卷烟风格特征。[Objective]To study the quantitative analysis method of six flavonoids in Apocynum venetum extractum,investigate the transfer behavior of the main functional components of A.venetum extractum,and its effects on the sensory quality of cigarettes and the reduction of free radicals.[Method]Using 20 mL 70%methanol-water as the solvent,ACQUITY UPLC HSS C_(18)(2.1 mm×100 mm,1.8μm)as the separation column,quercetin-D5 as the internal standard,and ultrasonic extraction-UPLC-MS/MS method was used to determine the content of six flavonoids in A.venetum extractum.The effects of different ingredient ratios on the sensory quality and harm reduction of cigarettes,as well as the transfer behavior of the main functional components of A.venetum extractum were studied.[Result]The UPLC-MS/MS method could achieve rapid separation of six flavonoids within 10 minutes,with a good linear relationship(R^(2)>0.9990)in the standard working curve.The LOD and LOQ of the method were 0.39-0.76 and 1.29-2.55 ng/mL,respectively.The recovery rate was 93.11%-105.77%,and the RSD was 0.73%-2.89%.Adding A.venetum extractum could improve the aroma quality of cigarettes and reduce smoke irritation;the transfer rate of quercetin in mainstream smoke was 0.89%-2.93%,and the maximum gas-phase free radical scavenging rate could reach 30%.[Conclusion]This method is easy to operate,has fast response,high accuracy and good repeatability,and can be applied to the rapid determination of active ingredients in A.venetum extractum.The application results of cigarette additives show that A.venetum extractum can effectively inhibit the release of free radicals in mainstream cigarette smoke.The appropriate application ratio(≤1.0%)can effectively improve the sensory quality of blended cigarettes and maintain the style characteristics of cigarettes.
