水红花子醇提物对大鼠肝纤维化及血清细胞因子水平的影响

摘  要:目的探讨水红花子醇提物对大鼠肝纤维化和血清细胞因子水平的影响。方法选取Wistar大鼠40只,随机分为对照组、模型组、低剂量组、中剂量组和高剂量组,每组8只。除对照组外,其余4组分别于大鼠腹腔注射猪血清0.5 ml,2次/周,连续12周;低剂量组、中剂量组和高剂量组分别灌胃药物0.58 g/(kg·d)、1.16 g/(kg·d)和2.32 g/(kg·d),1 ml/100 g,1次/d,连续12周。采用酶联免疫吸附法(enzyme linked immunosorbent assay,ELISA)检测大鼠血清透明质酸(hyaluronic acid,HA)、层黏连蛋白(laminin,LN)、Ⅳ型胶原(type IV collagen,Ⅳ-C)和Ⅲ型前胶原(procollagenⅢ,PCⅢ)含量。采用Masson染色观察肝组织纤维化改变。采用Luminex xMAP检测大鼠血浆细胞因子水平。结果与对照组相比,模型组HA、LN、IV-C及PCⅢ明显升高;3种不同剂量药物灌胃后,与模型组相比,中剂量组HA、LN、IV-C及PCⅢ明显下降,差异有统计学意义(P<0.05)。Masson染色结果显示,模型组可见点状坏死和慢性炎细胞浸润、肝索排列紊乱,纤维结缔组织增生,肝小叶结构破坏,提示造模成功;不同剂量药物灌胃后,低剂量组和中剂量组纤维间隔变薄、胶原沉积减少、肝小叶趋于完整,其中以中剂量组改善更为显著。与对照组相比,模型组Th1类细胞因子IL-2、IFN-γ的水平均下降;Th2类细胞因子IL-4、IL-6、IL-13和IL-5均升高;粒细胞集落刺激因子(G-CSF)和调节激活正常T细胞表达分泌因子(RANTES)均升高,差异有统计学意义(P<0.05)。与模型组相比,中剂量组IL-2、IFN-γ的水平均升高,Th2类细胞因子IL-4、IL-6、IL-13和IL-5均下降,G-CSF、RANTES均下降,差异有统计学意义(P<0.05)。结论水红花子醇提物能降低大鼠血清HA、LN、Ⅳ-C及PCⅢ含量,调节大鼠血清细胞因子分泌水平,通过改善免疫功能,发挥对大鼠肝纤维化的抑制作用。Objective To explore the effects of fructus ploygoni orientalis(FPO)on liver fibrosis and serum cytokine levels in rats.Methods A total of 40 Wistar rats were selected,and were randomly divided into control group,model group,low-dose group,medium-dose group,and high-dose group,with eight rats in each group.Except for the control group,the other four groups were injected with 0.5 ml of pig serum intraperitoneally,twice a week for 12 weeks.The low-dose group,medium-dose group,and high-dose group were given drugs 0.58 g/(kg·d)、1.16 g/(kg·d)and 2.32 g/(kg·d),1 ml/100 g by gavage,respectviely once a day for 12 weeks.Levels of serum hyaluronic acid(HA),laminin(LN),type Ⅳ collagen(Ⅳ-C)and procollagen Ⅲ(PCⅢ)were detected by enzyme linked immunosorbent assay(ELISA).The changes of hepatic fibrosis were observed by Masson staining.Luminex xMAP was used to detect the level of plasma cytokines in rats.Results Compared with the control group,HA,LN,Ⅳ-C and PCⅢ in the model group increased significantly,while compared with the model group,HA,LN,Ⅳ-C and PCⅢ in the medium-dose group decreased significantly after three different doses of drugs given by gavage,with statistically significant differences(P<0.05).Masson staining results showed that punctate necrosis and chronic inflammatory cell infiltration,disordered arrangement of hepatic cords,proliferation of fibrous connective tissue and destruction of hepatic lobule structure were observed in the model group,which suggested that the model was successful.After different doses of drugs given by gavage,the fibrous septa became thinner,collagen deposition decreased and hepatic lobules tended to be complete in the low-dose group and the middle-dose group,especially in the middle-dose group.Compared with the control group,the levels of Th1 cytokines IL-2 and IFN-γ decreased,while the levels of Th2 cytokines IL-4,IL-6,IL-13 and IL-5 all increased,both granulocyte colony-stimulating factor(G-CSF)and secretory factor that regulate and activate that expression of nor
