摘 要:为筛选烟草黑胫病生防菌并提高生防菌防效,从烟草根际土壤中分离筛选高效生防菌,通过形态学、生理生化和分子生物学方法对其进行鉴定,利用单因素试验优化生防菌的发酵条件,并对其发酵液与氟噻唑吡乙酮复配后对黑胫病的防效进行初步评价。结果表明:筛选到的拮抗菌株CY2经鉴定为多粘类芽孢杆菌(Paenibacillus polymyxa),对烟草疫霉的平板对峙抑菌率为82.1%,且具有广谱抑菌能力;CY2最适发酵培养基为NB培养基,可溶性淀粉和酵母粉分别为最适碳源和氮源,优化后的发酵条件为初始pH 8、30℃、180 r/min、发酵时长36 h、接种量2.0%,发酵液菌量增加到9.2×10^(8)cfu/mL,抑菌率达93.5%;当CY2发酵液与氟噻唑吡乙酮按照1∶2的体积比复配后,与单独使用氟噻唑吡乙酮的防效无显著性差异,防效均超过78%,但复配剂的施用可有效减少化学农药的使用量,为后续CY2发酵液复配氟噻唑吡乙酮的田间应用提供重要参考。To screen biocontrol bacteria against tobacco black shank disease and to improve the efficacy of biocontrol bacteria,bacterial strain CY2 with excellent antagonistic activity against Phytophthora parasitica was isolated from rhizosphere soil of healthy tobacco plants.The strain CY2 was identified by morphological,physiological,biochemical and molecular biological methods and its fermentation conditions were optimized by single factor experiments.Preliminary evaluations were made on the disease control efficacy of the combined CY2 fermentation broth and oxathiapiprolin against black shank disease.The results showed that strain CY2 was identified as Paenibacillus polymyxa and its inhibition rate against P.parasitica was 82.1%in the plate confrontation test with a broad spectrum of antifungal activities.The optimal fermentation medium for strain CY2 was NB medium with soluble starch and yeast extract as carbon and nitrogen sources,respectively.Under the optimized fermentation conditions,namely initial pH 8,fermented for 36 h at 30℃with shaker speed of 180 r/min and 2.0%inoculum size,the inhibition rate of the fermentation broth reached 93.5%when the bacterial amount increased to 9.2×10^(8)cfu/mL.When CY2 fermentation broth was combined with oxathiapiprolin at a 1∶2 volume ratio,there was no significant difference in the control efficacy compared to that of using oxathiapiprolin alone,with both exceeding 78%efficacy.However,the application of the combined agents could effectively reduce the amount of chemical pesticides used.These results provide an important reference for the subsequent field application of CY2 fermentation broth combined with oxathiapiprolin.
