竹节参总皂苷对小鼠急性毒性实验的研究

摘  要:目的观察竹节参总皂苷对小鼠的急性毒性,为竹节参的后续研究提供基础的实验数据,为临床用药剂量选择和剂型开发等相关研究提供参考。方法通过测定最大耐受量,初步判断竹节参总皂苷对小鼠的急性毒性或安全范围。将健康成年昆明小鼠随机分为对照组和试验组,每组10只,雌雄各半,每天1次连续灌胃5天;对照组给予0.5%羧甲基纤维素钠溶液,试验组分别给予竹节参总皂苷的3个剂量(500mg/kg/d、1500mg/kg/d、4500mg/kg/d);观察小鼠外观、行为、对刺激的反应、分泌物、排泄物、体重变化、死亡情况等。5天后对所有小鼠进行大体解剖,检查小鼠脏器病变情况。结果与对照组相比,试验组小鼠无中毒症状和异常活动反应,体重、进食量、对刺激的反应、分泌物、排泄物、各脏器指数指标均无显著差异。结论小鼠对竹节参总皂苷最大耐受量大于4500mg/kg/d,竹节参皂苷具有较高的安全性。Objective To test the acute toxicity of Total Saponins of Panax Japonicum(TSPJ)in Mice,to provide basic experimental data for the subsequent study of Panacis japonici rhizome,and to provide references for the related studies on the selection of clinical dosage and the development of dosage form.Methods The acute toxicity of TSPJ was observed by maximum tolerance dose(MTD)method.Kunming mice were randomly divided into four groups:control group and drug administration groups,with 10 mice in each group,half male and half female.For 5 days,the control group was given 0.5%CMC-Na solution by gavage,and the experimental groups were given three doses of TSPJ(500 mg/kg/d,1500 mg/kg/d,4500 mg/kg/d),the mice were continuously observed for 5 days to record their general state,body mass,food consumption,response to stimuli,secretions,excreta,and appearance of major organs,as well as their poisoning and death.Results In the acute toxicity experiment,no poisoning or death was observed in mice in each dose groups.There was no significant difference in the general condition,body mass,food consumption,response to stimuli,secretions,excreta,and appearance of major organsand organ index of mice before and after administration.Conclusion The MTD of TSPJ in mice is greater than 4500mg/kg/d,indicating that TSPJ has less toxic side effects and good safety.
