摘 要:目的采用高效液相色谱(HPLC)结合多元统计分析,建立定喘汤HPLC指纹图谱及含量测定方法。方法采用Waters SPHERISORB ODS2(250 mm×4.0 mm,5μm)色谱柱,以体积分数0.2%磷酸水溶液(B)-乙腈(A)进行梯度洗脱,流速1.0 ml/min,检测波长254 nm,进样量10μl,柱温35℃。以黄芩苷为参照,绘制10批样品的HPLC指纹图谱,对其相似度进行评价,确定共有峰;运用统计学软件进行多元统计分析,同时对定喘汤中6种有效成分的含量进行测定。结果10批定喘汤共标定了17个共有峰,相似度值为0.925~0.996,指认出绿原酸、芦丁、野黄芩苷、异绿原酸C、黄芩苷和甘草素6个共有峰。聚类分析和主成分分析结果基本一致,均将10批样品分为3大类。绿原酸、芦丁、野黄芩苷、异绿原酸C、黄芩苷、甘草素的质量浓度分别在0.010~0.166、0.005~0.084、0.021~0.339、0.016~0.250、0.082~0.824和0.005~0.086 mg/ml内与各自峰面积呈良好的线性关系,平均回收率范围为96.7%~101.0%,相对标准偏差(RSD)均不大于1.7%(n=9),测得10批定喘汤中绿原酸、芦丁、野黄芩苷、异绿原酸C、黄芩苷、甘草素含量分别为3.683~4.150、1.363~1.580、6.079~6.897、4.783~5.296、23.46~26.53、1.860~2.283 mg/g。结论本研究建立的结合多元统计分析的定喘汤HPLC指纹图谱及含量测定方法简便,重复性好,适用于定喘汤的质量评价。Objective To establish the high-performance liquid chromatography(HPLC)fingerprint and the content determination method of Dingchuan Decoction by combined with multivariate statistical analysis.Methods Waters SPHERISORB ODS2(250 mm×4.0 mm,5μm)column was used for gradient elution with 0.2%phosphoric acid aqueous solution(B)-acetonitrile(A),the flow rate was 1.0 ml/min,the detection wavelength was 254 nm,the sample volume was 10μl,and the column temperature was 35℃.Taking baicalin as a reference,the HPLC fingerprints of 10 batches of samples were drawn,and their similarity was evaluated to determine the common peaks.Multivariate statistical analysis was carried out by using statistical software,and the contents of six effective components in Dingchuan Decoction were determined.Results There are 17 common peaks in 10 batches of Dingchuan Decoction,and the similarity value is 0.925~0.996.Six common peaks of chlorogenic acid,rutin,scutellarin,isochlorogenic acid C,baicalin and liquiritigenin were identified.The cluster analysis and principal component analysis results were basically consistent,and 10 batches of samples are divided into 3 categories.The mass concentrations of chlorogenic acid,rutin,scutellarin,isochlorogenic acid C,baicalin and liquiritigenin were in the range of 0.010-0.166,0.005-0.084,0.021-0.339,0.016-0.250,0.082-0.824 and 0.005-0.086 mg/ml,respectively,and showed good linear relationships with their respective peak areas.The average recovery rate ranged from 96.7%to 101.0%,and the relative standard deviation(RSD)were all less than 1.7%(n=9).The contents of chlorogenic acid,rutin,scutellarin,isochlorogenic acid C,baicalin and liquiritigenin in the 10 batches of Dingchuan decoction were 3.683-4.150,1.363-1.580,6.079-6.897,4.783-5.296,23.46-26.53 and 1.860-2.283 mg/g,respectively.Conclusion The HPLC fingerprint and content determination method of Dingchuan Decoction combined with multivariate statistical analysis established in this study is simple,reproducible,and suitable for the quality evalua
