摘 要:目的 确定珍珠透骨草中槲皮素最佳提取工艺。方法 以槲皮素提取率为指标,通过单因素试验筛选影响槲皮素提取率较大的三个因素,采用中心组合试验设计和响应面分析法,选出最佳提取工艺。结果 液料比、乙醇浓度、提取温度对珍珠透骨草槲皮素提取率的影响显著;最佳提取工艺为液料比21.151 mL·g^(-1)、乙醇浓度51.784%、提取温度50.898℃,在此条件下,模型预测的槲皮素提取率为9.763 mg·g^(-1);多次试验证实,该工艺提取得到槲皮素提取率为9.895 mg·g^(-1),比预测值高。结论 利用响应面分析法优化得到的珍珠透骨草槲皮素的提取工艺可靠,可行性高。OBJECTIVE To determine the optimal extraction process of quercetin from Speranskia tuberculata.METHODS With the quercetin as the index,three factors affecting the extraction rate of quercetin were screened by a single factor experiment.The central combination experimental design and response surface methodology were used to select the optimal extraction process.RESULTS The best extraction process conditions of quercetin were follows:the liquid material ratio was 21.151 mL·g^(-1),ethanol concentration was 51.784%,ultrasonic temperature was 50.898℃.Under this extraction condition,the predicted extraction rate of quercetin was 9.763 mg·g^(-1).Confirmed by multiple trials,the extraction rate of quercetin was 9.895 mg·g^(-1).CONCLUSION The extraction process of quercetin from Speranskia tuberculata optimized by response surface methodology was reliable and feasible.
