摘  要:目的探索动脉粥样硬化动物模型的造模及验证方法。方法以8只6周龄的C57BL/6J小鼠作为对照组,8只6周龄的雄性ApoE-/-C57BL/6J小鼠作为模型组制备动脉粥样硬化(atherosclerosis,AS)模型,模型组与对照组均采用高糖高脂饲料连续喂养12~18周,分别于第12周和第18周进行解剖取材后行HE染色观察小鼠腹主动脉内膜厚度及斑块面积。结果与对照组相比,模型组小鼠12周后开始出现明显的动脉内膜增厚及脂质沉积,并随着时间的延长逐渐从脂纹期进入纤维斑块期,甚至粥瘤期,镜下明显可见动脉内膜增厚,胆固醇结晶及纤维帽形成(P<0.05)。结论ApoE-/-C57BL/6J小鼠在高糖高脂饲料连续喂养的情况下可成功诱导AS模型形成,通过对小鼠腹主动脉取材,利用HE染色后镜下观察可明确AS模型是否成功。Objective To explore the modeling and validation methods of animal models of atherosclerosis.Methods 6-week-old C57BL/6J mice as the control group,6-week-old male ApoE-/-C57BL/6J mice as the experimental group for the atherosclerosis model.Both the experimental group and the blank group were continuously fed with high-sugar and high-fat diet for 12-18 weeks.The abdominal aortic intimal thickness and plaque area were observed by HE staining at the 12th and 18th week,respectively.Results Compared with the control group,the experimental group began to show obvious arterial intima thickening and lipid deposition after 12 weeks,and gradually entered the fibrous plaque and even porridge tumor stage with the prolongation of time.Under the microscope,the arterial intima thickening,cholesterol crystallization and fibrous cap formation were obvious.Conclusion ApoE-/-C57BL/6J mice can induce the formation of AS model under the condition of continuous feeding with high glucose and fat diet.The success of AS model can be determined by sampling the abdominal aorta of mice and observing under the microscope after HE staining.
