摘  要:对广西某鸭场临床疑似由新型鸭呼肠孤病毒(Novel duck reovirus,NDRV)感染的雏鸭进行RT-PCR鉴定、NDRV分离、毒株细胞培养特性、雏鸭致病性、S1基因序列测定和遗传进化分析等研究。结果显示,RT-PCR检测组织病料为NDRV阳性,组织样品处理后接种SPF鸭胚,鸭胚出现发育迟缓、死亡,胚体全身及多脏器出血等症状;用鸭胚尿囊液接种雏鸭后出现脾脏坏死的特异性病变。S1基因的同源性与进化树分析结果显示,GX2022株与公布的部分NDRV毒株核苷酸序列相似性为93.7%~98.9%,而与鸭呼肠孤病毒(Muscovy duck reovirus,MDRV)GX2010株、GX110058株、S1133株,与禽呼肠孤病毒(Avian reovirus,ARV)815株、ZJM2000M株同源性很低,在21.8%~28.4%之间。进化树分析显示,该毒株与我国流行的NDRV处在同一个分支上,与MDRV和ARV的距离较远,说明分离到的毒株是新型鸭呼肠孤病毒。毒株接种LMH、CEF、Vero细胞后均产生空泡和细胞崩解等细胞病变。该新型鸭呼肠孤病毒临床感染的致病特征研究,有助于新型鸭呼肠病毒病的防控及疫苗的研发。This study conducted the RT-PCR identification,duck embryo isolation virus and cell culture characteristics,pathogenicity of ducklings,S1 gene sequencing,and genetic evolution analysis on a clinically suspected case of ducklings infected with the novel duck reovirus(NDRV)in a duck farm in Guangxi.The results showed that the specific RT-PCR detection of tissue samples was positive for NDRV.After inoculating SPF duck embryos with the tissue samples,it caused delayed development and death of the embryos,as well as bleeding from the entire body and multiple organs of the embryos;Inoculation of ducklings resulted in specific lesions of spleen necrosis.The homology and evolutionary tree analysis of S1 gene showed that the nucleotide sequence similarity between GX2022 strain and some published NDRV strains was 93.7%-98.9%.However,the homologies with duck reovirus(MDRV)GX2010 strain,GX110058 strain,S1133 strain,and avian reovirus(ARV)815 strain and ZJM2000M strain are very low,ranging from 21.8%to 28.4%.The evolutionary tree showed that the virus was on the same branch as the prevalent NDRV in China,but was further away from duck reovirus(MDRV)and avian reovirus(ARV),This indicates that the strain we isolated is a novel duck reovirus.The strains inoculated into LMH,CEF,and Vero cells all produced cellular lesions such as vacuoles and cell disintegration.The results of this study provided technical references for the prevention and control of novel duck reovirus disease and the development of vaccines.
