摘 要:仓储害虫的为害是造成粮食储藏损耗的主要因素之一,其体内微生物与害虫生物学特性密切相关。为探究锈赤扁谷盗、长角扁谷盗、烟草甲、锯谷盗和印度谷螟5种常见仓储害虫体内微生物的种类与群落结构,利用高通量测序技术对害虫体内微生物16S rRNA的V3-V4变异区进行测序,分析害虫体内微生物的多样性。结果显示,5种仓储害虫共得到原始序列1941590条,有效序列1814302条,2040个OTUs(Operational Taxonomic Unit,操作分类单元),注释到41个门、72个纲、142个目、219个科、370个属,其中Apibacter属为近缘物种锈赤扁谷盗和长角扁谷盗体内的优势菌属,沃尔巴克氏体属(Wolbachia)为烟草甲和锯谷盗的优势菌属,芽孢杆菌属(Bacillus)为印度谷螟的优势菌属。根据Alpha多样性分析,印度谷螟和锈赤扁谷盗的微生物多样性比长角扁谷盗、烟草甲和锯谷盗高。通过主坐标分析与聚类树两种Beta多样性分析,研究了五种害虫间体内微生物的相似性,结果表明不同物种的体内微生物存在显著性差异,但害虫亲缘关系愈疏远,微生物群落相似性越低。研究结果表明物种是影响昆虫体内微生物多样性的重要因素之一,且物种的亲缘关系越近,其微生物群落相似性越高,本研究为仓储害虫和微生物的互作研究奠定基础。Stored products pest is one of the primary factors contributing to grain losses,and their microbiota is closely tied to their biological characteristics.To investigate the types and community structures of the gut microbiota in five common species of stored products pests,including Cryptolestes ferrugineus,Cryptolestes pusillus,Lasioderma serricorne,Oryzaephilussurinamensis and Plodia interpunctella,16S rRNA V3-V4 variable regions were sequenced using high-throughput sequencing to study the diversity of their microbiota.A total of 1941590 raw tags,1814302 effective tags and 2040 operational taxonomic units(OTUs)were generated in the five species of insects.These OTUs were categorized into 41 phyla,72 classes,142 orders,219 families and 370 genera.The Apibacter was prominently dominant in two closely related species C.ferrugineus and C.pusillus,while Wolbachia dominated in L.serricorne and O.surinamensis.Bacillus was the dominant genus in P.interpunctella.The Alpha diversity analysis revealed that the microbial diversity of P.interpunctella and C.ferrugineus was higher than that of C.pusillus,L.serricorne and O.surinamensis.By using PCoA and clustering tree analysis of Beta diversity analysis,the similarity of microbial communities of five pests was studied.The results indicated that there were significant differences in microbial communities among different species,but the more distant the genetic relationships between pests,the lower the similarity of microbial communities.The results indicated that species played a crucial role in microbial diversity,with related species exhibiting greater similarity in microbial communities.This study established a foundation for exploring the interactions between stored products pests and their associated microorganisms.
