摘 要:在脂肪酸合成过程中,3-酮酯酰-ACP合酶Ⅱ(KASⅡ)对长链脂肪酸的合成起关键作用。本研究依据转录组数据设计特异性引物,从澳洲坚果果仁中克隆得到1个MiKASⅡ基因,并对其进行生物信息学分析。结果表明,澳洲坚果果仁MiKASⅡ长456 bp,可编码152个氨基酸,利用生物信息学预测,Mi KASⅡ蛋白质分子式为C_(713)H_(1141)N_(201)O_(214)S_(7),蛋白分子量为16.2 kD,理论等电点为6.50,被预测为亲水性蛋白,蛋白中不存在跨膜结构域,蛋白结构主要由α折叠、延伸链、β-转角和无规则卷曲组成。经与其他14种植物进行系统发育树分析,结果显示,澳洲坚果的MiKASⅡ与蒜头果的聚为一支,亲缘关系相对较近。实验克隆了澳洲坚果的MiKASⅡ基因,并对MiKASⅡ基因的结构、性质等进行预测,有利于深入研究该蛋白在脂肪酸的生物合成和代谢过程中的调控机制。3-ketoacyl-ACP synthaseⅡ(KASⅡ)plays a key role in the synthesis of long chain fatty acids in the process of fatty acid synthesis.A MiKASⅡgene was cloned from Macadamia nut kernel by designing specific primers based on transcriptome data,and its bioinformatics analysis was carried out.The results showed that the length of Mi KASⅡwas 456 bp,encoding 152 amino acids.Bioinformatics prediction showed that the molecular formula of MiKASII protein was C_(713)H_(1141)N_(201)O_(214)S_(7),the molecular weight of the protein was 16.2 kD,the theoretical isoelectric point (p I)was 6.50.MiKASⅡ was a hydrophilic protein without transmembrane domain,and the protein structure was mainly composed of α-sheet,extended chain,β-turn and irregular curl.Phylogenetic tree analysis with other 14 plants showed that MiKASⅡof Macadamia and KASⅡof Malania oleifera were clustered into one branch,and the genetic relationship was relatively close.In this study,the cloning of Mi KASⅡgene and its structure and properties are helpful to further study the regulatory mechanism of MiKASⅡprotein in the biosynthesis and metabolism of fatty acids in Macadamia.
