摘  要:根据前期筛选到的阳春砂成花途径差异基因CONSTANS(CO)设计特异引物,通过PCR克隆获得基因全长CDS序列,利用NCBI进行同源序列比对分析确认为AvGhd7基因,其全长CDS为693 bp,编码230个氨基酸残基,含有CO基因典型的CCT结构域,属于CCT超基因家族。系统进化分析发现AvGhd7与姜科植物聚为一类。基因表达分析表明,阳春砂AvGhd7基因在开花部位的表达量显著低于其它组织部位,在早实单株中的表达量显著低于非早实单株,为阳春砂开花性状的负调控基因。本研究可为阳春砂早实性状的分子机制研究提供参考。The study employed specific primers derived from differentially expressed genes CONSTANS (CO)previously identified within the flowering pathway of Amomum villosum.to amplify and clone the full-length coding sequences (CDS) of these genes.Homology sequence comparison analysis via NCBI revealed that one such gene is AvGhd7,which has a full-length CDS of 693 bp encoding 230 amino acid residues.This gene contains the characteristic CCT domain associated with CO and belongs to the CCT supergene family.Phylogenetic analysis indicates that AvGhd7 clusters closely with ginger species.Furthermore,gene expression analysis showed that the expression level of AvGhd7 gene in flowering site was significantly lower than that in other tissues,and the expression level of AvGhd7 gene in early-bearing individual plant was lower than that in non-early-bearing individual plant suggesting it functions as a negative regulator of flowering in Amomum villosum.This research provides valuable insights into the molecular mechanisms underlying early fruiting traits in Amomum villosum.
