摘 要:本研究旨在应用非靶向代谢组学方法,探究不同比赛途程伊犁马赛前血浆代谢组差异,筛选不同比赛途程伊犁马潜在标志差异代谢物。以参加伊犁马常态化赛事2000 m和80 km比赛伊犁马为研究对象,在比赛前采集所有马匹颈静脉血液制备血浆,在年度比赛结束后根据比赛成绩,各选取2000 m和80 km比赛成绩最佳的6匹马的血浆样本,采用液相色谱-质谱(LC-MS)对血浆样本进行非靶向代谢组学检测,筛选两组样本间的差异代谢物,并进行通路富集分析。结果表明:在两组马匹间共筛选出110种差异代谢物,与80 km组马匹相比,2000 m组马匹检测出56种显著上调差异代谢物,54种显著下调差异代谢物。差异代谢物显著富集到精氨酸和脯氨酸代谢、生物素代谢等代谢通路,涉及的主要差异代谢物为L-鸟氨酸、肌酐、肌酸和庚二酸。由此可见,筛选出的差异代谢物可能可以作为不同比赛途程伊犁马早期运动选材参考指标,为后续深入探讨伊犁马健康养殖和专项训练方案制定提供数据参考。This study aimed to apply non-targeted metabolomics methods to explore the differences in plasma metabolites of Yili horses before the race at different distances,and to screen for potential marker differential metabolites of Yili horses at different race distances.Yili horses participating in the regular 2000 m and 80 km races were selected as the research objects.Before the race,blood samples were collected from all horses'jugular veins to prepare plasma.After the annual races,the plasma samples of the top 6 horses with the best performance in the 2000 m and 80 km races were selected based on their race results.Liquid chromatography-mass spectrometry(LC-MS)was used to detect the plasma samples in a non-targeted metabolomics approach,screening for differential metabolites between the two groups of samples,and conducting pathway enrichment analysis.The results showed that a total of 110 differential metabolites were screened out between the two groups of horses.Compared with the 80 km group,the 2000 m group of horses detected 56 significantly up-regulated differential metabolites and 54 significantly down-regulated differential metabolites.The differential metabolites were significantly enriched in metabolic pathways such as arginine and proline metabolism,and biotin metabolism,with the main differential metabolites being L-ornithine,creatinine,creatine,and adipic acid.Therefore,the screened differential metabolites may serve as reference indicators for early selection of Yili horses at different race distances,providing data reference for further exploration of Yili horse health breeding and specialized training program development.
