摘 要:为优化四味穿心莲散半仿生提取物(SBSCS)的制备工艺,本试验利用DPS19.05版软件进行3因素5水平的均匀试验设计测定各组浸膏率,以微孔-平板法测定SBSCS体外对猪大肠杆菌的最小抑菌浓度(MIC),高效液相色谱法(HPLC)检测SBSCS中绿原酸、芦丁含量,流动相梯度洗脱液为乙腈-0.1%磷酸水;以浸膏率、MIC、绿原酸及芦丁含量组成的综合评分为响应值,以提取溶媒pH、料液比、提取时间为因变量,优化分析SBSCS制备工艺参数。结果显示:SBSCS制备最优的工艺为pH 6.8、料液比1:20、提取时间2.0 h。此条件下,浸膏率为(37.81±0.12)%、MIC为12.5 mg/mL、绿原酸含量为(85.22±0.23)μg/mL、芦丁含量为(3.12±0.26)μg/mL。该项研究获得了制备SBSCS的最优工艺,为其作用机制及新产品研发奠定了基础。The purpose of this study was to obtain the preparation technology of semi-biomimetic extract on Siwei Chuanxinlian San(SBSCS).The extractum yield of each group was determined by the uniform test design of 3 factors and 5 levels using DPS19.05 software.The minimum inhibitory concentration(MIC)of the each group extract against porcine E.coli were determined by microporous plate method in vitro.Acetonitrile-0.1%phosphoric acid water as mobile phase gradient elution,and the contents of chlorogenic acid and rutin in the each group extract were detected by high performance liquid chromatography(HPLC).The comprehensive scores composed of extract rate,MIC,content of chlorogenic acid and rutin were used as response values.And the extraction process parameters were optimized with solvent pH,solid-liquid ratio and extraction time as dependent variables.The results showed that the optimal preparation process of SBSCS was pH 6.8,solid-liquid ratio 1:20,extraction time 2.0 h.Under this experimental conditions,the extract yield was(37.81±0.12)%,MIC against porcine E.coli was 12.5 mg/mL,chlorogenic acid content was(85.22±0.23)μg/mL,rutin content was(3.12±0.26)μg/mL.This study obtained the optimal SBSCS,which laid a foundation for its mechanism of action and the development of new products.
