摘 要:丝素蛋白支架材料在组织工程领域中极具应用前景,其力学性能对组织再生修复中细胞的增殖、分化及生长过程具有至关重要的影响。文章以CaCl_2-甲酸为溶剂溶解脱胶蚕丝后,采用盐析法制备丝素蛋白支架是一种有效且简便的方法,所制备的支架材料具有较高的压缩模量和强度,但由于高质量分数甲酸对丝素蛋白具有显著的促结晶作用,反向的低压缩模量和强度调控并不易实现。对此,文章通过对丝素蛋白甲酸溶解液进行加湿处理,稀释溶解液中甲酸的质量分数,实现了对丝素蛋白支架材料力学性能的反向调控。研究结果表明,引入加湿处理环节后,所制备的丝素蛋白支架材料具有良好稳定的成形性,孔隙率约90%,孔壁由丝素蛋白微球组成,压缩模量和强度随着加湿处理时间的增加而逐渐降低,有效实现了其力学性能可调控性,期望能够促进适用于不同组织再生丝素蛋白支架材料的设计制备。Silk fibroin,which is derived from natural silk,is a renewable resource renowned for its excellent biocompatibility,non-toxicity,low immunogenicity,ease of functional modification,and good structural tunability.It is widely utilized in the regeneration of various tissues,including skin,blood vessels,ligaments,cartilage,bone,and nerves,as well as in clinical research.As a crucial material for biomedical tissue engineering,silk fibroin scaffolds feature an interconnected three-dimensional network structure,suitable plasticity,and favorable mechanical properties.These scaffolds play an essential role in supporting cell proliferation,differentiation,and adhesion.An effective and straightforward method for preparing silk fibroin scaffolds involves dissolving degummed silk in a solution of CaCl 2-formic acid,followed by a salt-leaching process.The resulting scaffolds exhibit high compressive modulus and strength;however,achieving lower compressive modulus and strength can be challenging due to the significant crystallization-promoting effect of high-concentration formic acid on silk fibroin.To achieve reverse modulation of the mechanical properties of silk fibroin scaffolds prepared using the salt-leaching method with a CaCl 2-formic acid solvent system,this study designed and implemented a custom humidification treatment apparatus and protocol.By applying humidification to the silk fibroin formic acid solution,the concentration of formic acid in the solution was diluted,enabling the reverse regulation of the mechanical properties of the silk fibroin scaffold materials.During this process,the effects of varying humidification treatment times and different amounts of porogen(sodium chloride)on the structure and properties of the prepared silk fibroin scaffold materials were investigated.The characterization of the prepared silk fibroin scaffold materials was conducted using scanning electron microscopy(SEM),Fourier-transform infrared spectroscopy(FTIR),X-ray diffraction(XRD),thermal analysis(TGA-DSC),mechanical property
